Objective: This study is aimed to investigate the cardioprotective effects of sulodexide pretreatment against myocardial ischemia/reperfusion injury in Sprague-Dawley rats.Methods: Select healthy,male,adult Sprague Dawley rats,while dividing them randomly into three groups: SO group,I/R group and VDF group.Build the myocardial ischemia/reperfusion injury model in SD rats.Electric physiological instrument was used to assess ECG.After the pericardium was opened,a silk suture was placed around the anterior descendant branch of the left anterior descending(LAD)coronary artery.The LAD was only exposed but not ligated in SO group,I/R groups were applied with 30 min of coronary artery occlusion and 120 mins reperfusion.Sulodexide was administered 15mg/kg.d i.g in 7 days before experiment.Myocardial infarct size was measured using EB/TTC staining method.Blood samples were collected for measurement CK-MB,TNF-?,IL-6,SOD and MOA concentrition.All calculations were computed with the aid of SPSS statistical software(version 20.0).All clinical datas were analyzed with SPSS20.0 version.P Value of less than 0.05(2-tailed)was considered to be statistically significant.Results:1 Comparison of baseline SD rats characteristicsAll experimental animals were male SD rats,250-350 g weight,10-12 weeks age old and SPF level,which were provided by experimental house of the first hospital affiliated to Northern Institute of Hebei Province.64 rats were selected in this experiment,with 4 rats died because of ventricular fibrillation occurred after ligation LAD in the process of building models.Among them,three rats died in I/R group and one in VDF group.Finally,60 rats were enrolled and 20 in with each group,while each?and?group had10 rats separately.2 ST-T Changes in electrocardiogram in ratsBefore the experiment,ECG was roughly normal when rats were in quiet condition.After successful ligation in I/R and VDF groups,ECG showed large abnormal QRS,ST segment arched-shape elevation.After reperfusion,T wave dropped quickly,with ST segment dropped down more than 50%.I/R group had 3 rats dead because of ventricular fibrillation,5 rats had short ventricular tachycardia.In VDF group,1 rat occurred ventricular fibrillation,4 had ventricular tachycardia.3 Comparison of myocardial infarct areaRats in?group accpted myocardial tissue EB/TTC staining.After dyeing,normal regional myocardial infarction in rats were visible blue,while myocardial ischemia area was dark red,myocardial infarction area was pale white.Contrast was obvious.Use the Image Pro Plus Image software to analyze infarction area,calculate the proportion of infarct area size and dangerous area size(ARS)(%).Results showed that,compared with I/R group,Sulodexide pretreatment could redece infarct size significantly(58.23±8.06% vs.45.16±7.91%,P<0.05).4 Comparison of myocardial biomarkers in SD ratsCompared with SO group,the levels of CK-MB in I/R group and VDF groups were elevated significantly(P<0.05);while levels in VDF groups were lower than those in I/R groups(3284.89±302.41 vs.1912.45±238.07U/L,P<0.05).5 Comparison of TNF-?and IL-6 in SD ratsTNF-?levels in SO group,I/R group,VDF group were 37.52±13.90Pg/ml,201.79±26.29 Pg/ml,144.13±35.58 Pg/ml separately.IL-6 levels in each group were 59.75±10.84 Pg/ml,174.15±18.58 Pg/ml and 132.27±11.03Pg/ml.Compared with SO group,levels of LDH and CK-MB in I/R group and VDF groups were elevated significantly(P<0.05);while levels in VDFgroups were lower than those in I/R groups(P<0.05).6 Comparison of SOD and MDA in SD ratsCompared with SO group,the concentrations of SOD in I/R group and VDF groups were lower(P<0.05);while levels in VDF groups were higher than those in I/R groups(69.80±17.23 vs.90.76±10.19 U/mg Prot U/L,P<0.05);Compared with SO group,the concentrations of MDA in I/R group and VDF groups were lower(P<0.05);levels in VDF groups were higher than those in I/R groups(7.35±1.38 vs.4.04±0.97 nmol/mg Prot,P<0.05).Conclusions:1 Sulodexide pretreatment can exert cardioprotective effects against myocardial ischemia/reperfusion injury in SD Rats,reduce myocardial infarct size,reduce myocardial biomarkers concentrations like LDH and CK-MB,and avoid the damage on cell structure.2 Sulodexide pretreatment can effectively suppress cell inflammation reaction in rats,decrease formation and release of inflammatory factors like TNF-? and IL-6.3 Sulodexide pretreatment can remove oxygen free radicals in rats,protect myocardium.
