Objective:SD rats as the research object of this subject, based colorectal mechanical stimulation and maternal separation principle prepared SD neonatal rat model of irritable bowel syndrome with IBS-D animal model.With the IBS visceral hypersensitivity closely related to BDNF-TrKB signaling pathway as a starting point to study Tongxieyaofang and its components to ease relations IB S visceral hypersensitivity and BDNF-TrKB signaling pathway between the cent ral dogma of gene transcriptions depth Tongxieyaofang square mitigation princi ple of IBS visceral hypersensitivity.Methods:Model group, SD rats 8-21 days after birth were accepted PTC A balloon dilation of the colon (pressure 60mmHg)In addition, stroked the perin eum of normal rats, given adequate food and water.120 rats were randomly sel ected 10 female rats and 10 male rats were used as normal rats.SD rat pups separated from their dams 1 hour (modeling time included) d aily. When grown to 8 weeks, through the colon expansion method to detect visceral sensitivity of each, the rats abdominal withdrawal reactions (AWR:abd ominal withdrawal reflex).All rats except the normal rats were randomly divide d into four groups:model group, the Tongxieyaofang group, Baishaofangfeng g roup, Chenpibaizhu groups.The next day to be fed with the drug treatment.The y were fed once a day, for 28 consecutive days.During the experiment, each rat was observed body hair situation, mental state, activity, responsiveness to external stimuli, stool.Before and after treatmen t and modeling each rat were weighed. Meanwhile, in the above-mentioned per iod of time to detect each rat stool water content and Bristol Stool Scale score. Detected each rat AWR score before and after treatmentThe next day after the end of treatment all rats were sacrificed to collect their blood, removal of the colon from the anus 2-5cm long and hippocampus brain tissue.Use the hippocampal tissue and colonic epithelial tissue to extract RNA, extracting all genes BDNF signals from the chip system includes raw data (BD NF, TrkB, etc.) the table values.Results:? Rats general observation:The normal group rats:their body hai r neat, mental status and activity is good, flexible response, normal stool grain y.The model group rats:3 days after modeling began to appear untidy hair, lis tlessness malaise, reduced activity levels, decreased ability to respond, the grow th rate decreased, increased number of particles in the stool, perianal hair in lo ose stools contaminated.The Tongxieyaofang group and Baishaofangfeng group r ats:the gradual emergence of mental state improved, enhanced activity, respons iveness to rise gradually from the unformed stool was normal to semi-formed t o particulate gradually easefed after fed drug 3-5 days; diarrhea basic relief, w eight gain speed gradually afte fed drug 10-14 days.After 10 days of drug tre atment, mental status and activity in rats slightly improved ability to respond t o a gradual recovery, weight gain rate increased slightly compared with the mo del rats, stool improved. But to the end of the experiment, rats were still a s mall part of the case of partial thin stool.? The amount of Increase weight in rats:compared with the normal group, the amount of weight increase was significantly less than the control group(P <0.05). After treatment, the Tongxieyaofang group and Baishaofangfeng group h ad higher weight gain than the model group, there was significant difference (P<0.05); The Chenpibaizhu group had no significant effect on body weight o f rats (P> 0.05).? Observed the situation of rats:After modeling, After model, compared with the normal group, the model group rats'stool water content and Bristol s tool score were significantly higher than the normal group (P<0.05;After treat ment, compared with the model group, the Tongxieyaofang group and Baishaof angfeng group rats' stool water content and Bristol stool score significantly lo wer (P<0.05); Compared to the control treatment itself,the tongxieyaofang grou p and the Baishaofangfeng group rats' stool water content and stool Bristol sto ol score lowered(P<0.05); while the model group and the Chenpibaizhu group rat' stool water content and stool Bristol stool score had not significant differ ence in the period(P> 0.05).? Visceral pain threshold:Compared with the normal group, the visceral p ain threshold of the model group were relatively low, visceral hypersensitivity(P <0.05);Visceral pain threshold of male and female rats with the same period of comparison, the female rats visceral pain threshold is significantly lower tha n male rats visceral pain threshold(P<0.05);After treatment, the visceral pain t hreshold of the Tongxieyaofang group and Baishaofangfeng group compared wit h before treatment increased, statistically significant differences (P<0.05);Before and after the treatment of visceral pain threshold in the modol group and the Chenpibaizhu group rats with no significant change, the difference was not st atistically significant (P> 0.05).? Pathology specimens of SD rats:The colonic epithelium and hippocamp us of each gruop rats did not see significant inflammatory cell infiltration and interstitial edema, pathological changes in line with IBS.? Gene expression of BDNF-TrkB in rats:BDNF mRNA and TrkB mRN A in each group of rats highly expressed in the hippocampus, the correspondin g low expression in the presence of colon tissue;Further, the expression of Trk B mRNA in colon tissue and hippocampus were significantly higher than BDN FmRNA;In colon tissue, BDNF mRNA and TrkB mRNA expression values in t he model group was significantly higher than the normal group(P<0.05);BDNF mRNA and TrkB mRNA expression value is lower than the model group in t he Tongxieyaofang group and the Baishaofangfeng group (P<0.05);In the hippo campus, BDNF mRNA and TrkB mRNA expression values in the model group were lower than normal, while the hippocampus TrkBmRNA expression values in the model group increased compared with the normal group;TrkBmRNA ex pression value in hippocampus compared with the model group no significant d ifference in the Tongxieyaofang group,the Baishaofangfeng group and the Ch enpibaizhu group,but there is a clear downward trend.Conclusion:(1) Expansion eight days old of rats'colon with PTC A ballo on (expansion pressure of 60mmHg), can be successfully established rat model of IBS-D.(2) Tongxieyaofang can improve the pathological state in the model group of IBS-D.(3) The "yimu"-Baishaofangfeng components of Tongxieyaofang is an activ e ingredient in the treatment of IBS-D.(4) BDNF-TrkB signaling pathway may be one of the mechanisms of IBS visceral hypersensitivity formation.(5) The Baishaofangfeng components of Tongxieyaofang relieve IBS-D lax ative side of visceral hypersensitivity possibly by BDNF-TrkB signaling pathwa y to achieve, so as to achieve remission IBS-D abdominal pain, diarrhea and o ther symptoms.
