In this study, podophyllotoxin is identified as a typical anti-breast cancer in vitro by MTT assay on the basis of four human breast cancer cells MCF7, MDA-MB-231, MDA-MB-453 and SK-BR-3. Analysis and comparison of podophyllotoxin on breast cancer cells in each human IC50 value, confirmed the podophyllotoxin sensitive cell lines MDA-MB-453 and SK-BR-3. Hoechst staining and flow cytometry are used to detect the ability of podophyllotoxin to induce HER2-positive breast cancer cell apoptosis. Western blot and real-time quantitative PCR are used to detect the expression of Cleaved-caspase-3, Cleaved-caspase-9, Bcl-2, Bax and other apoptosis-related proteins and mRNA expression levels. The results show that compared with 5FU and emodin, the inhibitory effect of podophyllotoxin on proliferation of human breast cancer cells is more significant (P<0.05), and could be used as a typical anti-breast cancer natural medicine in vitro. Compared to HER2-negative breast cancer cells MCF7 and MDA-MB-231, both HER2-positive breast cancer cells MDA-MB-453 and SK-BR-3 cells underwent podophyllotoxin-induced apoptosis (P<0.05). The mechanism may be through the activation of Cleaved-caspase-3, Cleaved-caspase-9 and up-regulation of Bax, down-regulation of Bcl-2 activation of mitochondrial pathway to induce HER2-positive breast cancer cell apoptosis (P<0.05).
