Cancer,one of the most lethal diseases,is serious harmful to people's lives.It's very necessary to find out the effective prevention and treatment for the incidence increased year by year.Hepatocellular carcinoma(HCC)is the most common primary malignant tumor of the liver,which has the characteristics of occult,high degree of malignancy,rapid progression,strong invasion and metastasis,and poor prognosis.Invasion and metastasis are the main reason of influencing the prognosis of malignant tumor,tumor cells through epithelial-mesenchymal transition(EMT)acquire invasive and metastatic potency.Therefore,it can effectively inhibit the invasion and metastasis of hepatocellular carcinoma,that inhibition of epithelial mesenchymal transition.This subject is to investigate Schisandrin A,Schisandrin B and Schisandrol A inhibited hepatocellular carcinoma the epithelial-mesenchymal transition.In the study of Schisandrin A,Schisandrin B and Schisandrol A inhibited hepatocellular carcinoma EMT.MTT assay show that the proliferation of hepatocellular carcinoma cells was inhibited by Schisandrin A,Schisandrin B and Schisandrol A in a concentration-dependent manner.Wound Healing and Transwell invasion assay respectively show that Schisandrin A,Schisandrin B and Schisandrol A have good ability to inhibit cancer cell migration and invasion.We detected the change of E-cadherin and N-cadherin,molecular markers of epithelial mesenchymal transformation by high-content screening,we found that SchisandrinA,SchisandrinB and SchisandrolA increased E-cadherin levels and decreased N-cadherin protein expression.Colony forming test demonstrated the single-cell proliferative capability of liver cancer cells were significantly inhibited.luciferase reporter assay was tested for transcription factors associated with cancer,such as Snail 1?Snail 2?Twist 1 and Twist 2.We found that SchisandrinA,SchisandrinB and Schisandrol A have significant function of regulating expression of transcription factors,Schisandrin A,Schisandrin B and Schisandrol A were verified anti-tumor mechanisms at the molecular level.This study found that Schisandrin A,Schisandrin B and Schisandrol A have good antitumor activity,have great potential value of anti-tumor.Multiple experiments proved that Schisandrin A,Schisandrin B and Schisandrol A reverse epithelial mesenchymal transition and suppresse the proliferation and metastasis of hepatocellular carcinoma cells.
