Malus hupehensis,is a general term for many kinds of plants of the apple genus(Malus)and papaya(Chaenomeles),which were cultivated in many parts of our country.This article has mainly studied the extraction,chemical properties,structure,antioxidant and anticancer activity of extraction from Malus hupehensis.Hope to lay the foundation for extraction of Malus hupehensis industrialized development and new drug discovery on cancer therapy.The extraction of the solvent was carried out at 80 ? for 2 times.Combined the two extracts and concentrated under reduced pressure.The crystals were verified by infrared spectroscopy.The antioxidant activities of extraction from leaves of Malus hupehensis were investigated including scavenging activity of hydrogen radicals,ABTS radicals,hydroxyl radicals and DPPH radicals in vitro.The anti-tumor activity of phloridzin was verified by MTT assay,activated oxygen(ROS),TUNEL,JC-1 and immunofluorescence.The results showed that we had established the water extraction technology for phytocyanin from Malus hupehensis and got the final product of phloridzin.The phlorizin had certain scavenging effect on various free radicals(antioxidant activitie)and the scavenging effect is the order of sABTS free radical> DPPH radical> Hydroxyl radical> metal ion Fe2+free radicals,and the antioxidant ability has a certain concentration-dependent,which lay a good foundation for the study of antioxidant activity.The phloridzin had a certain cytotoxicity on the ovarian cancer cells Skov3 and A2780,and Hela cells.Compared with the control group,the fluorescence intensity about ROS was significantly higher in the treated group of Skov3 cells.Meanwhile,the extraction resulted in nuclear DNA leakage,accompanying histone ?-H2 AX and RAD51 foci,which eventually lead to apoptosis.It was shown that the extraction of Malus hupehensis leaves has antitumor activity.This study provides basic data for the widespread use of phlorizin for antitumor research and treatment.
