?Objective?To detect the differentiation change of osteopontin-related Lnc RNAs in sodium oxalate crystallization induced renal damages via bioinformatics approach,and to discuss the mechanism of osteopontin-related Lnc RNAs in renal injuries.?Methods?Renal injury model was established by stimulating renal tubular epithelial cell with sodium oxalate.Osteopontin expression level in in vitro model of renal tubular epithelial cell(HK-2)stimulated by sodium oxalate was detected by RT-PCR.Osteopontin knock-down cell model was carried out by RNA interference.Human genome lnc RNA microarray(V3.0)offered by Arraystar Co,Ltd was utilized in the detection of osteopontin-related Lnc RNAs and m RNAs differentially expressed in renal tubular epithelial cell crystallization models of 3 si-osteopontin groups and 3 control groups.lnc RNAs was derived from authoritative public transcriptome database(including Refseq?UCSCknowngenes?Gencode and other databases).Screening criteria of differential gene was set as Fold Change>1.5 and P<0.05.Gene ontology(GO),KEGG pathway analysis and other bioinformatics approaches were applied to the analysis of differentially expressed genes.?Results?1.Renal injury model was established by stimulating renal tubular epithelial cell with sodium oxalate,and osteopontin-interference model established by RNA interference technique.2.Human genome lnc RNA microarray indicated osteopontin-related Lnc RNAs expression in human renal injury model of renal tubular epithelial cell induced by sodium oxalate crystallization.There were a total of 583 genes of lnc RNAs with differential expression,including 354 up-regulation genes and 229 down-regulation genes in interference group;235 genes of m RNAs differential expression,including 139 up-regulation genes and 96 down-regulation genes.3.Gene ontology(GO),and KEGG pathway analysis results indicated lnc RNAs of differential expression was correlated to cell growth,enzyme activity regulation,PI3k/Akt/NF-?B and Wnt signaling pathways.KEGG pathway analysis indicated up-regulated lnc RNAs were related to 14 pathways,while down-regulated lnc RNAs were related to 5 pathways.Generalized analysis implied significantly gathering of NOD-like receptor signaling pathway,PI3k/Akt/NF-?B,Wnt signaling pathways and other inflammation-related pathways.?Conclusions?There were osteopontin-related Lnc RNAs expression in human renal injury model of renal tubular epithelial cell induced by sodium oxalate crystallization in vitro.Osteopontin-related Lnc RNAs were involved in the mechanism of the sodium oxalate crystallization induced renal damages,which can be used as biological indicator candidate providing theoretical and experimental evidence for renal injuries induced by crystallization.
