| OBJECTIVE:To observe the levels of thyroid autoantibodies(TGAb,TPOAb)and cytokines TNF-α and IL-10 in experimental autoimmune thyro-iditis(EAT)rats with Qishao Granules mixture of Radix Astrag ali,Radix Paeoniae Alba,Radix Curcumae etc.,And to explore the effect of traditional Chinese medicine mixture on EAT rats,and to provide a reliable theoretical basis for the clinical treatment of Hashimoto’s thyroiditis.Method:Establishment of model: 60 SPF SD female rats were used,a ged 4-6 weeks,body weight 110 ± 10 g.Transported to the Hubei University of Traditional Chinese Medicine Experimental Center room adaptive feeding for 1 week,and then the rats were rando mly divided into normal control group according to the quality of the body,and the rest was made of the model group.The mice in the normal control group were given distilled water at the second week,while the mice in the model group began drinking high iodine water at a concentration of 0.64 g/L.The mice in the model group were minutely injected into the water-filled TG (CF: CFA = 1: 1)by complete Freund’s adjuvant(CFA)The middl einterval of two days,each containing TG100μg,as the primaryimmune.At the end of the fifth week,TG was emulsified by inco mplete Freund’s adjuvant(IFA)at a posterior subcutaneous dose of 100 μg,once a week for 4 weeks,as a booster immunization.At the end of the 8th week,The rats were randomly divided into model group(n = 15),triptolide group(n = 15)and traditional Chinese medicine group(n = 15).The equivalent dose of the rat was calculated according to the equivalent dose ratio table of human and animal body surface area.Qishao Granules group per rat intragastric volume of 21.4g/kg/d.The rats of Tripterygium wilfordii polyglycidium group were treated with Tripterygium wi lfordii polyglycoside,and the water was mixed with 6.25mg/kg/d.The model group and the normal control group were fed with the same amount of distilled water.4 groups were given once a day for 4 weeks.The rats were sacrificed at the 12 th week,and the levels of serum TGAb,TPOAb,TNF-α and IL-10 were measured by enzyme-linked immunosorbent assay(ELISA).Result: 1.The changes of TGAb and TPOAb antibodies in each group: the c hanges of TGAb and TPOAb antibodies in the Model group were sig-nificantly higher than those in the normal control group,and there were significant differences between the two groups(P <0.01);two antibodies of Qishao Granules group and Tripterygium g roup were significantly lower than those in the model group,an d model group is significant the difference(P<0.01);there is no significant difference between the Qishao Granules group and Tripterygium group(P>0.05);2.The changes of cytokine levels in each group:The change of TNF-level:the level of alpha TNF-alpha in the model group was significantly higher than that in the normal control group,there was significant difference compared with the normal control group(P<0.01);The levels of TNF-α in Qishao Granules group and Tripterygium group were significantly lower than that in the model group,compared with model group were significantly different(P<0.01);There was no significant difference in the levels of TNF-α between the Qishao Granules group and the triptolide group(P> 0.05).The change of IL-10 level:the level of IL-10 in model grou p was significantly lower than that in the normal control group,there was significant difference compared with the normal contr ol group(P<0.01);The levels of IL-10 in the Qishao Granules group and Tripterygium group were significantly higher than those in the model group,compared with model group were signif-icantly different(P<0.01);there is no significant difference between Qishao Granules group and Tripterygium group(P>0.05).Conclusion: 1.TGAb and TPOAb in the model group were significantly higher than those in the normal control group,The experimental model of autoimmune thyroiditis(EAT)was successful.2.Qishao Granules and tripterygium glycosides tablets can effec t-tively reduce the levels of TGAb and TPOAb antibodies in thyroid autoantibody of EAT rats,and there is no significant difference between Qishao Granules and Tripterygium wilfordii polyglycoside tablets.3.Both Qishao Granules and tripterygium wilfordii polyglycoside could significantly decrease serum TNF-α and increase the level of IL-10 in EAT rats.There was no significant differencebetween the two groups in reducing the level of serum TNF-α and IL-10 in EAT rats.Compared with Tripterygium wilfordii polyglycoside tablets,there was no significant difference.4.Qishao Granules and Tripterygium wilfordii polyglycoside effect on the intervention effect of EAT rats. |
