Endoplasmic reticulum stress(ERS)plays an important role in type two diabetes,ERS can promote the apoptosis of pancreatic beta cells,so it is of great theoretical significance to study the mechanism between ERS and pancreatic beta cells in the prevention of type two diabetes.Antibacterial peptide AWRK6(SWVGKHGKK FGLKKHKKH)is a new antibacterial peptide obtained from the optimization of the laboratory.The preliminary experimental results show that antibacterial peptide AWRK6 plays a role in cell proliferation and insulin secretion,and peptide AWRK6 in the body is not easy to be degraded,is expected to further develop to become efficient and safe for the treatment of type two diabetes drug.The experimental results showed that the antibacterial peptide AWRK6 can inhibit the apoptosis of pancreatic beta cells induced by endoplasmic reticulum stress.In order to explore the mechanism of antibacterial peptide AWRK6 inhibits the apoptosis function,based on the synthetic antibacterial peptide AWRK6 and mouse islet cells as experimental object,using MTT assay,Western blot and other experimental techniques,a preliminary analysis on the mechanism of antibacterial peptide AWRK6 inhibits apoptosis of Min6 cells caused by endoplasmic reticulum stress.Using MTT colorimetric assay for cell survival rate,Min6 cells using tunicamycin(induced by endoplasmic reticulum stress,Tunicamycin)were treated according to the experimental results show that the antibacterial peptide AWRK6 at the concentration of 100nmol/L the cell survival rate of normal group and TM group were significantly different(P<0.01),so the subsequent experiments were performed using 100nmol/ the concentration of antibacterial peptide AWRK6.The Western blot results showed that Western blot,has induced endoplasmic reticulum stress in MIN6 cells after AWRK6 treatment effect of Epac2 expression was increased,endoplasmic reticulum stress molecules Chop and Bip expression decreased,Bax decreased expression of Pro apoptotic molecules,and joined the GLP-1 receptor inhibitor,the inhibitory effect of antimicrobial peptide AWRK6 on endoplasmic reticulum stress and apoptosis was inhibited to some extent.In order to investigate the hypoglycemic effect of AWRK6 in vivo and the effects of endoplasmic reticulum stress on apoptosis,a model of type two diabetic rats was established by streptozotocin(STZ).The results showed that the antibacterial effect of antimicrobial peptide AWRK6 was only in the condition of high glucose,and it did not cause hypoglycemia reaction.The results of OGTT experiment showed that the glucose tolerance of rats with type two diabetes treated with AWRK6 was significantly improved.Extraction of islet tissue protein for Western blot,also found that after AWRK6 treatment can make the effect of Epac2 expression was increased,endoplasmic reticulum stress molecules Chop and Bip expression decreased,Bax decreased expression of Pro apoptotic molecules.The results of this study illustrate the antibacterial peptide AWRK6 by improving the molecular mechanism of endoplasmic reticulum stress,inhibit the apoptosis of islet cells,for the further development of AWRK6 as a treatment for type two diabetes has laid a theoretical and experimental basis.
