Germinal centre(GC)is a kind of substructures which exists in peripheral lymphoid organs.Within GCs,B cell undergo a series of programs which include proliferation,somatic hypermutation,antigen dependent selection and finally differentiated to plasma cell or memory B cell.Although the basic processes and principles of germinal centre response have been disclosed,the detailed molecule mechanisms are still unknown.Here we first report that REGy,a 20S proteasomes activator,could regulate germinal centre response.We demonstrate that REGy deficient mice showed dramatic decrease of antigen specific antibody level in serum.After isolation of human tonsilar B cell subtypes,we find REGy specifically expressed in germinal centre B cells,rather than other cell types.REGy deficient mice show normal B cell development and comparable germinal centre B cell percentage in payer's patch.REGy is dispensable for germinal centre establishment and maintenance after NP-CGG immunization,but REGy deficiency impairs antibody secretion both in the first immunization and the second immunization.More than that,we also proved that REGy deficiency could protect mice from CIA.We speculate that this phenomenon is caused by dysfunction of plasma cell,but we do not know the molecular details or cannot exclude the possibility that other cell involved in this process.In summary,we demonstrate that REGy could regulate germinal centre B cell response and REGy deficiency would alleviate CIA in mice.
