Objective To investigate the effect of modified bone mesenchymal stem cells(BMSCs)with gene co-expression of bone morphogenetic protein 2(bmp2)and vascular endothelial growth factor 165(vegf165)on histomorphology and biomechanics of tendon-bone healing after rabbit anterior cruciate ligament reconstruction.Methods Sixty-six white rabbits were randomly divided into three groups: experimental group,control group,normal group.The tendon-bone interface was injected with BMSCs transfered by lentiviral vector with gene co-expression of bmp2 and vegf165 in experimental group,BMSCs without transfection in control group and none in normal group.Histomorphology and biomechanics were evaluated in 3rd,8th and 12 th weeks after surgery.Results Histological pathology revealed that neovascularization and fibroblast cells infiltration in experimental group were higher than that in the control group.Ability of tissue healing was better than the control group owing to the formation of typical 4-layered tendon-bone interface in 12 th week postoperatively.Biomechanics test found that the maximum loads in the experimental group showed significant superior outcomes compared with the control group(P<0.05).Conclusion The modified BMSCs with gene co-expression of bmp2 and vegf165 may improve the outcome of tendon-bone healing.
