Objective:Myeloid-derived suppressor cells(MDSCs)are a heterogeneous population of immature myeloid cells that suppress innate and adaptive immunity.MDSCs are present in many disease settings;However,at present,the study of MDSCs are more concentrated on the field of tumor,Tumor and host cells in the tumor microenviron-ment(TME)produce a myriad of pro-inflammatory mediators that utilize a variety of mechanisms to suppress T cell activation,induce other immune-suppressive cell populations,regulate inflammation in the TME,and promote the switching of the immune system to one that immune escape and enhances tumor growth.However the study of MDSCs in autoimmune disease,especially in autoimmune thyroid diseases,is rarely reported.This study is aim to investigate the expression of myeloid-derived suppressor cells(MDSCs)in the patients with Graves disease.Methods: A total of 52 newly diagnosed GD patients from January 2015 to September 2015,and 30 healthy people were enrolled in the study,which be divided into disease group(GD)and healthy controls,CD14+HLA-DRlow/-was used as molecular markers for MDSCs.The frequency of MDSCs in GD patients and healthy controls were determined by flow cytometry.ELISA was used for detecting the concentrations of plasma cytokines Arg-1?IL-6 and G-CSF.The correlation between the frequency of MDSCs and the function of thyroid was also analysed.According to the patient's compliance and signed informed consent of patients,we primarily choose23 patients with GD given anti-thyroid medication therapy.According to the test results to adjust the treatment dose of anti-thyroid drugs,Six months later,we detect MDSCs in peripheral blood using CD14+HLA-DRlow/-as molecular markers when patients condition are smmoothy.Results The frequency of MDSCs was significantly increased in GD patients compared with that in healthy controls.This frequency was also obviously decreased in GD patients when treated with anti-thyroid drugs(P<0.05).There was no significant correlation was found between the frequency of MDSCs and the function of thyroid.The concentrations of plasma cytokines IL-6 and G-CSF were significantly increased in comparison with that in the healthy controls,while this was not existed in Arg-1.Conclusion The increased frequency of MDSCs may play an important role in the occurrence and development of Graves disease.
