Relationship Between The Homocysteine Levels And CBS G919A Gene Polymorphisms And Intracranial Atherosclerotic Stenosis In The Patients With Ischemic Stroke

Objective:To investigate the relationship between plasma homocysteinemi levels,ge-ne polymorphisms of cystathionine beta-synthase(CBS)G919A and stenosis of intracranial and extracranial artery.Methods: In a retrospective case-control study,we collected patients with ischemic cerebrovascular disease 148 cases,according to the presenceof cerebral vascular sten-osis divided into 86 cases of cerebral vascular stenosis and 62 cases non-stenosis.The plasma Hcy levels were measured by fluorescence popar-ization immunoassay(FPIA),CBS G919 A gene polyorphisms were ananlyed PCR and restriction fragment length polymorphism(PCR-RFLP)and amplification refractory mutation system(AR MS).Results:(1)Hcy level of male in experimental group is higher than the control group(17.81 ± 7.3μmol/L,13.66 ±7.2μmol/L,t = 2.52,P < 0.01);the Hcy level of women in experi-mental group is also higher than the control group(13.35±4.1μmol/L,7.88±2.9 μmo l/L,t = 6.44,P < 0.05);And comparing the same group founds that the male Hcy con-centration is significantly higher than female Hcy levels in the experimental group and control group;and the difference had statistical significance connection.(2)The genotype homozygous mutant type AA,heterozygous mutation GA compared with wild type GG genotype has no statistical significance(X2= 0.196,P>0.05).The distribution of genotype and allele frequency does not show statistical difference in th-e experimental group and control group(X2= 0.219,P>0.05);(3)The variance analysis showed that plasma Hcy concentration was statistically sign-ificant between different genotypes of CBS G919 A including the wild type GG type and heterozygous mutation GA comparison: t = 5.89,P < 0.05;Wild type GG compared with homozygous mutant AA: t = 6.67,P < 0.05;Heterozygous mutant GA compared with homozygous mutant AA: t = 2.15,P < 0.01.Conclusion: High homocysteine levels are a risk factor for intracranial atherosclerotic stenosis;cystathionine beta-synthase(CBS)G919A exists gene polymorphism;The homozygous mutations(AA)and heterozygous mutations(GA)is one of the im-portant genetic factors that affect plasma Hcy levels;cystathionine beta-synthase(C BS)G919A gene polymorphism and intracranial atherosclerotic stenosis in a given area has no relevance.
