AimStudy whether the synergistic effect of pDNA-EDA and pDNA-EGF was stronger than the effect of single pDNA-EDA on the sweat glands regeneration.MethodsFirst,a gene-activated collagen-chitosan/silicone scaffold with bone mesenchymal stem cells(MSC)as seed cells was fabricated.The scaffold with pDNA-EDA was treated as control groups(group EDA),and the scaffold with pDNA-EDA and pDNA-EGF was treated as experiment groups(group EDA+EGF).SD rat models with full-thickness skin defects in the ventral foot skin were treated with the scaffold of each group.2 weeks,4 weeks and 8 weeks after transplantation,we general observed the wound healing of the feet skin.Moreover,we evaluated the regenerated tissues stained with hematoxylin and eosin microscopically.Finally,immunohistochemical staining of cytokeratin 8(CK8)and cytokeratin 14(CK14)was used to identify sweat gland.ResultsThere was no significant difference of wound repair between the control groups(group EDA)and experiment group(group EDA+EGF)been with the naked eye at 2 weeks,4 weeks and 8 weeks after transplantation.Two groups both healing well,and no obvious scar left at the 8th week after transplantation.HE staining analyze showed that the regenerated tissue in the experimental group(group EDA+EGF)was similar to normal skin tissue at 8 weeks,and a large amount of fat cells and the sweat-glands-like structures were found.At the same time,most of the cells in the control group(group EDA)were fibroblasts,the sweat-glands-like structures were also found,but the number was fewer and the structure was simple.Immunohistochemical analysis of regenerated tissues showed CK8 and CK14 positive,indicating that the regenerated structure was sweat gland.ConclusionThe synergistic effect of pDNA-EDA and pDNA-EGF was stronger than only pDNA-EDA in collagen-chitosan/silicone scaffold carrying MSC,to promote the repair of skin defects and induce rat sweat gland regeneration.
