BackgroundGastrointestinal tumors are a serious threat to human health, the latest ca magazine shows that in China, the incidence and mortality of gastric cancer are the highest in the second place in all tumors. Although with the rapid development of radiotherapy and immunotherapy, the survival of gastric cancer has been extended. However, because the discovery is not timely, most of the tumors are discovered in the late, making the overall survival rate of gastric cancer less than 30%. Therefore, early detection of gastric cancer ,the discovery of effective tumor markers , early diagnosis and early prevention and treatment are effective ways to improve survival. NOTUM gene is a newly discovered gene, has been confirmed in zebrafish ,it can be achieved by dephosphorylation to inhibit the wnt signaling pathway, and it finds in human liver cancer with high expression, and is detected as a role of negative feedback regulation in wnt signaling pathway. The study in gastrointestinal cancer is not clear, this study explores the expression of NOTUM gene in colon cancer and gastric cancer and its relationship with the clinical significance.ObjectiveTo detect the expression of NOTUM gene in gastric carcinoma and to explore its clinicopathological relationship with gastric cancer. To study the relationship between the expression of wnt signal pathway and NOTUM gene, to detect the expression of NOTUM gene in colorectal carcinoma and to explore its clinicopathological relationship with colon cancer. Further explore the role of NOTUM genes in digestive tract tumors.MethodsThe expression of NOTUM gene in tissues was detected by immunohistochemistry.The expression of NOTUM gene was correlated with the clinicopathological and prognosis of patients. The expression of NOTUM protein and ?-catenin protein was detected by immunoblotting. And the relationship between NOTUM protein and?-catenin protein was determined by SPSS correlation analysis. Kaplan-Meier method was used to compare the survival time of patients with gastric cancer and negative expression of gastric cancer. Log-rank test and Cox regression model were used to analyze the clinical and pathological features of prognosis.Results(1) The expression of NOTUM gene in gastric carcinoma was significantly increased, and its expression was correlated with lymph node metastasis and TNM stage and prognosis (p <0.05), but not with age, sex, tumor location, tumor size and tumor pathological type. There was a positive correlation between NOTUM gene and ?-catenin protein expression in gastric cancer tissues. The survival time of patients with positive expression of NOTUM gene was significantly shortened. NOTUM gene expression can be used as an independent risk factor for the survival of gastric cancer patients(2) The expression of NOTUM gene in colon cancer tissue was significantly increased, and its expression was correlated with lymph node metastasis and TNM stage and prognosis (p <0.05), which was not related to age, sex, tumor size and tumor pathological type. The survival time of patients with positive colon cancer with NOTUM gene expression was significantly shorter. NOTUM gene expression can be used as an independent risk factor for survival of colon cancer patientsConclusionNOTUM gene is expressed highly in the gastric cancer and colon cancer and can be used as a biological markers of the progression and prognosis in gastric cancer and colon cancer disease .
