Background:Flavonoid is a class of organic substances widely found in native plants.A large number of studies have shown that they have a wide range of biological activities,such as anti-tumor,anti-oxidation,anti-inflammatory,and so on.Hesperetin is a member of the family of dehydrogenate.And they a large number of legumes,butterflies,lilacidae and rutaceae,especially exist in a large number of rutaceae citrus plants peel and flesh.Studies have shown that hesperetin has the properties of dihydroflavonoids,for example,there are anti-tumor,anti-oxidant free radicals and other activities.Therefore,we speculate that hesperetin has anti-inflammatory immune regulation of biological activity.The function of the immune system is to prevent the body invaded by outside intrusion.Without the protection of the immune system,the body will be ready to be infected with unrelated antigens.But sometimes the immune system will generate the wrong answer,generating positive responses to self-antigens attack,autoimmune diseases;or the immune system to distant organs at the time of the transplanted organ rejection.Then we need immunosuppressive drugs to decrease the response of the immune system.Objective:This study was intended to use the mouse animal model to study the efficacy of anti-inflammatory hesperetin,hesperetin and immunoregulatory mechanisms during anti-inflammatory action.Method:1.To establish a model of mouse foots swelling.m BSA was once used in a sensitive state using an emulsifier formed by methylated bovine serum albumin and complete Freund’s adjuvant.And then use mBSA-PBS solution in the mouse foot subcutaneous injection,the next day can cause swelling of the mouse foot-swelling.The differences between the mice in the administration group and the mice in the control group and in the model group were important to note at the cell and molecular level.2.In vitro culture of mouse lymphocytes.In vitro control of differentconcentrations of hesperidin levels,found that the effect of hesperidin on lymphocytes,and the role of hesperitin on the cell concentration of the best concentration.3.High-throughput sequencing of long-chain non-coding RNA for immune regulation.The mechanism of anti-inflammatory effect of melatonin was determined and validated by high-throughput sequencing of long-chain non-coding RNA and a small amount of mRNA in foot-infiltrating lymphocytes of foot-swelling mass.Result:1.Hesperitin can alleviate the degree of foot-swelling in mice with sufficient swelling,decrease the release of proinflammatory cytokines in the blood,and thus play an anti-inflammatory role.2.Hesperitin can decrease the amount and ratio of lymphocytes in mice with sufficient swelling,and play an immunosuppressive role.3.Hesperitin in vitro lymphocyte proliferation experiments,in a relatively high concentration conditions can inhibit the proliferation of lymphocytes growth,with immunosuppressive effect.4.Hesperitin can change the regulation of cell cycle and govern the expression of cytokines such as lncRNA to affect lymphocyte apoptosis,inhibition of immune system function.Conclusion: Hesperitin has anti-inflammatory immunosuppressive function. |
