Object To analyze how penehyclidine hydrochloride protects lung with regard to ALI from observation of the impact on the expression of p38 mitogen-activated protein kinase(p38-MAPK)by pre-operating penehyclidine hydrochloride on the condition that lipopolysaccharide(LPS)caused ALI on rats.To offer new methods for prevention and cure of acute lung injury.Methods The reseach had two parts.The first part:thirty 7 days old Sprague-Dawley rats weighting 16-22 g were randomly divided into 3 groups(n=10 each).Acute lung injury was indused by intraperitoneal(IP)injection of lipopolysaccharide(LPS,5ml/kg)in Group B and Group C,.In Group C,rats were injected penehyclidine hydrochloride(2ml/kg)into the abdominal cavity half an hour before LPS administration.In Group A,rats received IP injection of normal saline(NS,5ml/kg).After 6 hours,we anaesthetized rats to collect the lung tissue to test the W/D and to observe the lung pathological features.We used ELISA to test TNF-? and IL-35 in lung,and used Immunohistochemical to study the expression of total p38-MAPK and p-p38-MAPK.The second part:forty-five 7 days old SD rats were randomly divided into 3 groups(n=15 each)as part 1.We observed the behavior changes and calculate the survival rate in 24 h of each group.Results The first part:Compared with Group A,the serum concentration of TNF-? and the W/D ratio were higher in Group B and Group C,while those in Group B were higher than Group C(P<0.05).The serum concentration of IL-35 was lower in Group B and Group C,and expressions of p38-MAPK and p-p38-MAPK protein were positive(P<0.05).The second part:2 hours after administration of intraperitoneal injection of LPS,rats bigan to tremble with polypnea and cold in group B.6 hours after establishing the rat model,the rats in group B began to appear shortness of breath,and even some of them have cyanosis.The symtoms of group C were lighter than group B.Rats in group A were normal in activity?reaction and eating.The rate of survivous in 24 hours of each group were 100%?66.7%?86.7%.Conclusions Pretreatment with penehyclidine hydrochloride has a protective action on LPS-induced acute lung injury of rats.The mechanisms of the effect may be through inhibiting the pathway of p38-MAPK.
