Malignant tumor is a public health problem that puzzles human life safety and quality of life.Melanoma,also known as malignant melanoma,is a type of skin cancer.Its incidence continues to increase,the mortality rate is very high,the situation is critical,and has become one of the major diseases that affect human health.The research of related vaccines has become the focus of researchers.Through the activation of the patient’s own immune system,the use of tumor cells or tumor antigens and induce specific cellular immunity and humoral immunity,enhance the body’s ability to prevent cancer,tumor growth,metastasis,proliferation and recurrence,in order to achieve the purpose of removal or control of the tumor.At present,tumor vaccines in clinical research tend to cellular immune response,while more effective vaccine should take into account the synergy between broad spectrum of antibodies and high levels of cellular immune.Toll like receptor 3 in natural killer cells,dendritic cells,CD4+T cells and other cells can express,and poly(I:C)binding,which makes it may become the research direction of polypeptide vaccine,a very valuable development.Poly(I:C)was given within 4 hours after peptide immunization,which could significantly improve the cellular immune responseMF59 adjuvant in all age groups,have good tolerance,can achieve long-term,cross protection,easy to produce,can induce the immune response to Th2.However,MF59 can be induced into Th1 reaction after the addition of pathogen associated molecular patterns and Toll like receptors.In this study,the tumor antigen survivin epitope peptide(68-81,95-104,80-88)were chosen as immunogen,supplemented with poly(I:C)and/or MF59 adjuvant to evaluate the immune effect and anti-melanoma activity.The results indicated that poly(I:C)and MF59 assisted survivin epitope peptide immunogen could control the tumor size,quality and volume of black melanoma mouse models.According to the analysis of antibody titer,antibody isotyping and ELISPOT,it is suggested that assisted immunogen can induce humoral immunity in mice.The results showed that poly(I:C)and MF59 combined with survivin95-104 could effectively induce humoral immunity mediated by Th2 cell(helper T cell 2).The study provides a basis for candidate immunogen design based on surviving,and provides support for that tumor therapy could induce a more balanced Th1/Th2 immune response.
