Objective:To investigate the inhibitory effect of combination of acitretin and interferon-a on leukemia model of mouse lymphoma.Method:1.Selecting C57 BL / 6 pure male mice,6-8 weeks old,weighing18-22 g.Leukemia cell line EL-4 was cultured in DMEM medium containing 20% horse serum.When the cells were grown to logarithmic phase,the cells were collected and formulated as1×107/ ml single cell suspension.2.Taking 0.1ml cells and inoculated on the dorsal side of the right hind limb in C57 BL / 6 mice,so the animal model was made.The administration was started on the second day after inoculation.3.Experimental grouping: Group A(blank control group):0.05 ml intraperitoneal injection of PBS,PBS solution 0.1ml gavage;Group B(high dose of Acitretin group): 0.05 ml intraperitoneal injection of PBS,Acitretin 20 mg / kg,0.1ml gavage;Group C(interferon group): 0.05 ml intraperitoneal injection of IFN?-2?,PBS solution 0.1ml gavage;Group D(combination group): 0.05 ml intraperitoneal injection of IFN?-2?,acitretin A20 mg / kg,0.1ml gavage;Group E(low dose Acitretin group): 0.05 ml intraperitoneal injection of PBS,acitretin A10 mg / kg,0.1ml gavage.The mice state and the tumor growth was observed daily.4.From the 6th day,the tumor volume and the weight of the mice were measured.The mice were executed on day 14,the tumor volume was measured by stripping tumor tissue and the tumor inhibition rate was determined by weight.Tissue fixed,sliced,HE stained.Measuring the levels of serum IL-6 by ELISA.Tumor weight,tumor volume,IL-6 cytokine expression and other measurement data with mean ± standard(x±s)deviation,The data were analyzed by SPSS 21.0 software.The results were expressed as x ± s.The mean value of each group was analyzed by LSD and P <0.05 for the difference was significant.Results:1.Mouse lymphoma leukemia model was established,the rate of tumor formation was100%.2.On the 7th day after inoculation,the antitumor effect of the drug was the strongest,and the antitumor effect was decreased on the 14 th day after inoculation.Acitretin can inhibit the proliferation of mouse T-cell lymphoma leukemia(p<0.05)IFN-? can inhibit the proliferation of murine T cell lymphoma leukemia(p <0.05),and Interferon and Acitretin combined with the most effective anti-tumor effect,the inhibition rate was 51.38%.3.HE staining showed abnormal cell division,no tumor metastasis was found.4.The results of ELISA showed that the concentration of IL-6in the experimental group was slightly lower than that in the negative control group,but there was no significant difference.Conclutions:1.In the early stage,Acitretin and interferon can inhibit the proliferation of mouse T cell lymphoma leukemia,and interferon? and Acitretin group of combined effect is the most obviously.2.IFN-?,Acitretin antitumor effect may be related to down-regulation of IL-6,the need for further experiments to improve.3.In the rapid growth period of the tumor,treatment with acitretin alone and / or interferon is not enough for the need for chemotherapy or other.
