Four Kinds Of Macromolecule-rich Traditional Chinese Medicine Injections (TCMI)Subacute Toxicity Experiments

Objective:Four kinds of Traditional Chinese Medicine Injections(TCMI)(Qingkailing injection,Danshen injection,Shuanghuanglian injection,Dengzhanxixin injection)rich in macromolecules were investigated to find toxic effect of the four TCMIs rich in macromolecules,thus to prove that macromolecular substance was the one of most important causes for TCMIs' adverse reaction.Methods:(1)The preparation of four kinds of injections rich in macromolecules(?10 k): Four kinds of injections rich in macromolecules were prepared with molecular sieve.Ten parts of injection was put in molecular sieves and spun in a centrifuge to intercept macromolecular substances above 10 k Dalton,at 4 ?,3000 rpm.1 part of injections rich in macromolecules was made.(2)Rabbits were randomly divided into blank control group,original injection group,and injection rich in macromolecule group.Rabbits' temperature was recoded with a ultra-red ear thermometer,and their weight was also measured,before and 2 hours after intravenous administration was taken.Intravenous administration was arranged once a day for 5 consecutive days,and temperature and weight were recorded.(3)After 5 day's intravenous administration,3 animals in every group were sacrificed,blood and sample and organs from the animals were collected.Routine blood test,blood biochemistry assay,organ coefficient,body temperature,weight,and pathological investigation were made.The remaining animals continued experimental study to observe recovery for additional 5 days without administration.Statistics analysis was made to find any difference between groups,or between before and after administration.Results:(1)The color of injections rich in macromolecules was obviously darker than that of the original injections.(2)The results of blood tests showed that some values of blood routine test and blood biochemical assay in injections rich in macromolecules groups increased to some extent.(3)In injections rich in macromolecules group,rabbit's weight decreased and body temperature increased to some extent.(4)By observing the pathological images of organs,some injury in injection rich in macromolecules group was able to be found,especially in liver and kidney tissue.The results suggested that,macromolecular substances were harmful for rabbit liver and kidney function.Conclusion:Injections rich in macromolecules had toxic effects on rabbits,and the macromolecules were one of the most important causes for TCMIs' adverse reaction.
