| Objective: Observation of Qi Yin granules on NAFLD rat liver tissue morphology and CYP2E1 protein expression in liver tissue,HOME-IR,Leptin,Resistin and IR related factor of insulin signal transduction components IRS-1,PI-3K mRNA,and discusses the mechanism of Qi Yin granules in the treatment of NAFLD.Methods: 72 male SD rats were randomly divided into blank control group,model group,low,middle and high dose group and positive drug control group,12 rats in each group.In addition to the control group,free eating and drinking,were given high fat emulsion by gavage to rats,1 times daily for 10 weeks,when the model was established successfully with Qi Yin particles of low medium and high dose of tiopronin and gavage treatment.To observe the pathological morphology of rat liver tissue after 20 weeks,the rats were measured the serum levels of Leptin,Resistin,fasting blood glucose and fasting insulin levels,and the calculation of HOME-IR,CYP2E1 protein content was detected by immunohistochemistry in the liver tissue,liver tissue content on the determination of IRS-1,PI-3K mRNA Real-time PCR.Results: Compared with the blank control group,model group rats serum Leptin,Resistin and HOME-IR were significantly increased,the content of IRS-1,PI-3K and mRNA in liver tissue of rats were significantly decreased,Leptin,and Resistin were increased in varying degrees,the difference was statistically significant(P<0.05),the NAFLD model successfully copied.Compared with the model group,the expression of CYP2E1 protein in each group,were significantly decreased the content of Resistin,Qi Yin granules in high dose group and positive drug control group Leptin and HOME-IR were decreased,the content of IRS-1,PI-3K and mRNA in liver tissue were significantly increased,the difference was statistically significant(P<0.05).Compared with the positive drug group,lower dose group Resistin content of Qi Yin particles,the content of IRS-1 and PI-3K mRNA in liver tissue were significantly increased,Qi Yin granules in high dose group Leptin,HOME-IR and CYP2E1 were decreased,the difference was statistically significant(P<0.05);compared with Qi Yin granules in low dose group,dose group Leptin Qi Yin particles decreased significantly,liver tissue IRS-1 and PI-3K of mRNA were significantly higher,Qi Yin particles in the high dose group of CYP2E1,Resistin and HOME-IR were decreased,the difference was statistically significant(P<0.05).Conclusion:(1)Qi Yin granule can reduce the protein expression level of CYP2E1,decrease the levels of Leptin,Resistin and HOME-IR in serum of rats,so as to achieve the purpose of improving IR and treating NAFLD.(2)Qi Yin granule can decrease the content of IRS-1 and PI-3K mRNA in the liver tissue of NAFLD rats,improve the activity of IRS-1 and PI-3K,reduce IR,and achieve the purpose of treating NAFLD. |
