Object: By observing the influence of Zhilong Huoxue Tongyu Capsule's tear open party component on the NF-kappa B pathway and inflammatory factor of atherosclerosis rats,Select its resistance AS main ingredients,in order to further clarify its mechanism of action,make the formula be streamlined.Methods :The 110 healthy male SD rats were divided into blank control group(n=10),model group(n=10),atorvastatin group(n=10),the whole party(n=10),decomposed recipe 1 group(n=10),decomposed recipe2group(n=10),decomposed recipe 3 group(n=10),decomposed recipe 4group(n=10),decomposed recipe 5 group(n=10),decomposed recipe6group(n=10),decomposed recipe 7 group(n=10)randomly.AS in the rat was established by the composite method of feeding high fat forage and intraperitoneal injections of vitamin D3 and immune damage,Groups of rats were feed for 120 d until the materials.In the building 60 days,we began to give the corresponding drug intervention,once-daily irrigation,a total of intervention for 60 days.The same volume of normal saline was taken to the blank control group and model group.In the 120 days of the experiments,preparing rat serum by take the blood of aortaventralis,and taking the rat thoracic aorta line HE staining and immunohistochemical staining.Using the method of biochemical to test the level of rats serum total cholesterol(TC),Triglyceride(TG),high-density lipoprotein cholesterol(HDL-C),low density lipoprotein cholesterol(LDL-C).Using the method of enzyme-linked immunoassay(ELISA)test the concentration in rat serum endothelin 1(ET-1),and Nitrate reduction method is used to determine the concentration of serum nitric oxide(NO)in rats.By using the immunohistochemical(IHC)method to measure the rat thoracic aorta positive expression of NF-kappa B p65,and using the HE staining method to observe aorta 's changes in the morphology and measure aorta's intima thickness in light microscope.Results:1.General situation of observation group rats: Blank control group rats were active flexible,eating good,hair luster;Model group rats were lazy,depression,eating significantly reduced,angular,hair lackluster.The drug intervention group rats were lively,sensitive reaction,the hair colour is normal,atorvastatin group and the whole party to eat well,the rest of decomposed recipe groups were eating a little reduce,but more food intake than model group.In the whole experiment process,there was no death in rats.2.The results of body weight changing of each rats group: Experiment to 30 d this period of time,the blank control group and model group rats weight has increased,model group rats weight grow faster than blank control group,The difference is statistically significant(p<0.05).The experiment for 30 d after 60 d this time,blank control group and model group rats weight growth rate has no obvious difference(P>0.05);From the experiment 60 d up to the end of the experiment,blank control group rats weight grow faster than the model group,The difference is statistically significant(p<0.05).The start to 60 d this period of time,model group rats weight with the drug intervention groups were increased,no difference in the growth rate(P>0.05).Experiment 60 d to 120 d this period of time,model group rats weight with the drug intervention group were increased,the drug intervention group rats weight growth rate was faster significantly than the model group(P < 0.05)3 According to the results of the orthogonal experiment of drugs on the AS rats serum TC,TG,LDL,HDL-C-C values :Intuitive analysis showed that,the primary and secondary order impact of herbs on serum TC value is: astragalus > cassia twig > lumbricus =leech >sargentgloryvine stem,The decomposed recipe which contain astragalus,cassia twig,lumbricus,leech reduce serum TC value.Variance analysis showed that,astragalus has an extremely significant influence on serum TC value(P<0.01),cassia twig,lumbricus,leech have a significant influence on serum TC value(P<0.05),sargentgloryvine stem has no significant effect on serum TC value(P> 0.05).Intuitive analysis showed that,the primary and secondary order impact of herbs on serum TG value is: astragalus > cassia twig > leech >lumbricus > sargentgloryvine stem.The decomposed recipe which contains astragalus,cassia twig,lumbricus and leech can reduce serum TG value.Variance analysis showed that,astragalus has an extremely significant influence on serum TG value(P<0.01),cassia twig,lumbricus,leech have a significant influence on serum TG value(P < 0.05),sargentgloryvine stem has no significant effect on serum TG value(P>0.05).Intuitive analysis showed that,the primary and secondary order impact of herbs on serum LDL-C value is:astragalus > cassia twig > lumbricus =leech > sargentgloryvine stem.The decomposed recipe which contains astragalus,cassia twig,lumbricus and leech can reduce serum LDL-C value.Variance analysis showed that,astragalus has an extremely significant influence on serum LDL-C value(P<0.01),cassia twig,lumbricus,leech have a significant influence on serum LDL-C value(P<0.05),sargentgloryvine stem has no significant effect on serum LDL-C value(P>0.05).Intuitive analysis showed that,the primary and secondary order impact of herbs on serum HDL-C value is: astragalus>cassia twig= leech>lumbricus > sargentgloryvine stem.The decomposed recipe which contains astragalus,cassia twig,lumbricus and leech can increase serum HDL-C value.Variance analysis showed that,astragalus,cassia twig,lumbricus,leech have an extremely significant influence on serum HDL-C value(P <0.01),sargentgloryvine stem has no significant effect on serum HDL-C value(P>0.05).4.According to the results of the orthogonal experiment of drugs on the AS rats serum ET-1,NO values : Intuitive analysis showed that,the primary and secondary order impact of herbs on serum ET-1 value is: astragalus>lumbricus>leech>cassia twig>sargentgloryvine stem.The decomposed recipe which contain astragalus,cassia twig,lumbricus,leech reduce serum ET-1value.Variance analysis showed that,astragalus,cassia twig,lumbricus,leech have a significant influence on serum ET-1 value(P<0.05),sargentgloryvine stem has no significant effect on serum ET-1 value(P>0.05).Intuitive analysis showed that,the primary and secondary order impact of herbs on serum NO value is: astragalus > cassia twig > leech > lumbricus >sargentgloryvine stem.The decomposed recipes which contain astragalus,cassia twig,lumbricus and leech can increase serum NO value.Variance analysis showed that,astragalus,cassia twig,lumbricus,leech have an extremely significant influence on serum NO value(P <0.01),sargentgloryvine stem has no significant effect on serum NO value(P>0.05).5.According to the results of the orthogonal experiment of drugs on AS rat thoracic aorta NF-kappa Bp65 positive expression: Intuitive analysis showed that,the primary and secondary order impact of herbs on the positive expression rate of NF-kappa Bp65 is: astragalus>cassia twig>lumbricus>leech>sargentgloryvine stem,the decomposed recipe which contain astragalus,cassia twig,lumbricus,leech can reduce the positive expression rate of NF-kappa Bp65.Variance analysis showed that,astragalus has an extremely significant influence on the positive expression rate of NF-kappa Bp65(P<0.01),cassia twig,lumbricus,leech have a significant influence on the positive expression rate of NF-kappa Bp65(P<0.05),sargentgloryvine stem has no significant effect on the positive expression rate of NF-kappa Bp65(P>0.05).6.According to the results of the orthogonal experiment of drugs on AS rat thoracic aortic intima thickness : Intuitive analysis showed that,the primary and secondary order impact of herbs on the thoracic aortic intima thickness is:astragalus > leech > cassia twig > lumbricus > sargentgloryvine stem,the decomposed recipe which contain astragalus,cassia twig,lumbricus,leech reduce the thoracic aortic intima thickness.Variance analysis showed that,astragalus,cassia twig,lumbricus and leech have a significant influence on the thoracic aortic intima thickness(P<0.05),sargentgloryvine stem stem has no significant effect on the thoracic aortic intima thickness(P>0.05).Conclusion:1.The primary and secondary order impact of herbs on the AS is: astragalus>leech>cassia twig>lumbricus>sargentgloryvine stem 2.The mechanism of Zhilong Huoxue Tongyu Capsule resist AS may be through the following ways:(1)Regulating lipid metabolism,decrease TC,TG,LDL-C,increase HDL-C to improve lipid metabolism disorders(2)Regulating vascular active substances,can increase the release of the release of NO and reduce ET-1 to protect the vascular endothelium?(3)NF-kappa B ‘s activity were inhibited to reduce the release of inflammatory cytokines.
