Objective: With the rapid development of most cities of our country,fog and haze increasingly prominent,quartz and nano-Si O2 are mainly two kinds of silicon rich fine particles of dust particles in haze weather.In this paper we select two kinds of silica rich mineral dust as the research object to explore the toxic effect on human lung adenocarcinoma cell line A549.Methods:(1)50 ?g/mL,100 ?g/mL,200 ?g/mL,400 ?g/mL,800 ?g/mL of two kinds of mineral dusts exposure to A549 cells for 24 h,the relationship between the survival rate and concentration of mineral dust were detected by MTT assay.(2)50 ?g/mL,200 ?g/mL and 800 ?g/mL of two kinds of mineral dusts exposure to A549 cells for 24 h,the activity of LDH was analyzed;200?g/mL of quartz and nano-Si O2 mineral dust suspension exposure to A549 cells for 3h,6h,18 h,then detect the changes of LDH activity.(3)50?g/mL,200 ?g/mL and 800 ?g/mL of mineral dust suspension exposure to A549 cells for 2 h,4 h,8 h,16 h,24 h,48 h,then detected the content of SOD and GSH.(4)The cell cycle and apoptosis rate of A549 cells was analyzed by flow cytometry with PI staining.Results:(1)Compared with the negative control group,with the increase of mineral dust suspension concentration,the survival rate of A549 cells decreased which showed a good dose-response relationship.(2)Compared with thenegative control group,the release amount of LDH which after A549 cells exposed to silica and nano-SiO2 mineral dust in suspension for 24 h was increased,and with the increase of mineral dust suspension concentration,the activity of LDH increased,there is a good dose-response relationship;the release of LDH of A549 cells induced by nano-Si O2 was higher than quartz.(3)The SOD activity of A549 cells was increased while the content of GSH was decreased with the increase of mineral dust concentration in a certain range of concentration and time.(4)Quartz and nano-Si O2 can induce A549 cell cycle arrest and apoptosis.Conclusion:Quartz and nano-SiO2 dust can destroy the A549 cell membrane,cause oxidative damage,cell cycle arrest and apoptosis,and the cytotoxicity of nano-SiO2 dust is more serious than in quartz dust.It is speculated that these two kinds of mineral dusts may affect the oxidation system,cell cycle and apoptosis of A549 cells in a certain concentration range.
