Objective:To explore the relationship among MUC1,integrin av?3 and RIF,and to analyze the predictive value of MUC1 and integrin av?3 for recurrent implantation failure by comparing the expression of MUC1 and integrin av?3 in the endometrium of the corpus luteum and the peripheral blood taken from women with recurrent implantation failure(RIF group),and women who have a history of pregnancy with IVF and would accept another cycle(control group),which was aim to provide theoretical basis for enhancing implantation rate.Methods:Patients who would receive the treatment of IVF-ET/ICSI-ET/Frozen-ET in the Department of Reproductive Medicine of the Second Affiliated Hospital in Shandong University of Traditional Chinese Medicine from June 2015 to July 2016 were selected.According to the inclusion criteria,the subjects would be divided into two groups.The patients failed to successfully pregnant who have more than three times history of embryo transfer with good-quality embryo or cumulative number of transplanted embryos more than 10 were RIF group in a total of 22 cases.The patients who have a history of pregnancy with IVF and would accept another cycle were selected as control group in a total of 24 cases.Intravenous venous blood and endometrial tissue were obtained at 7-9 days after LH peak.The expression levels of MUC1 and integrin av?3 in peripheral blood and endometrial tissues were detected by enzyme-linked immunosorbent assay(ELISA).Results:(1)The levels of MUC1 and integrin avP3 in the endometrial tissue and peripheral blood of RIF group were significantly lower than those of control group(P<0.05).(2)There was a positive correlation between the levels of peripheral blood-MUC1 and the expression of tissue-MUC1 in two groups(P<0.05).There was no significant correlation between peripheral blood-integrin av?3 and tissue-integrin av?3 in both groups(P>0.05)(3)There was a positive correlation between the levels of MUC1 and the level of integrin av?3 of both groups in peripheral blood(P<0.05),and there was a positive correlation between the levels of MUC1 and the level of integrin av?3 of both groups in the endometrium(P<0.05).(4)The area under the ROC curve(Area under Curve,AUC)of tissue-MUC1,tissue-integrin av?3 and peripheral blood-MUC1 were 0.955,0.885 and 0.879,respectively.The AUC of the three were greater than the area under the reference line(P<0.05).The optimal cut-off values for the three were 0.60 pg/mg,0.58 pg/mg,27.84 pg/ml,respectively.The sensitivity was 91.7%,70.8%and 75.0%,respectively.Conclusion:(1)The expression of MUC1 and integrin av?3 decreased,suggesting that the expression of local regulatory protein in endometrium of RIF decreased and the endometrial receptivity decreased,which may be one of the reasons for embryo implantation failure(2)The expression of MUC1 and integrin av?3 was relevant and may be regulated by some mechanism but the specific mechanism was subject to further study(3)Peripheral blood-MUC1 may serve as a noninvasive biological marker for assessing endometrial receptivity...
