Research purposes:Through vitro experiments,we investigate the possible mode of action of acupuncturing well point which controls Act A-mediated extracellular signal transduction through the neuronal Act A/Smads signal transduction pathway into the cells,and clarify the mechanism of action of acupuncture which controls activin A-mediated Act A/Smads signal pathway treat VD.Method:1.Make a model of vascular dementia rats.The rats were randomly divided into acupuncture group and model group,and the sham operation rats were used as the control group.2.By analyzing the experimental results of water maze and behavioral scores of rats to determine the success of rat dementia model.3.The morphological changes of hippocampal cells were observed by pathological sections.Adding the experimental group of serum to intervent culture after cultures of P12 cells,,the establishment of OGD model and blocking the activin A receptor,and finally detect each experimental group about Act A,Smad3 protein expression results.Result:1?Water maze test results show that compared with the model group,the escape latency of the acupuncture group was significantly shortened,and the number of times of the original platform was significantly increased after removing the platform.2?The results of cell morphology showed that compared with the model group,the apoptosis of hippocampal neurons in acupuncture treatment group was significantly decreased.3 ? The expression of Acta and Smad3 protein in the acupuncture serum intervention group was higher than that in the parallel culture group(P <0.05).The expression of Acta and Smad3 protein in the model serum intervention group was higher than that in the parallel culture group(P <0.05).The expression of Acta and Smad3 protein in the acupuncture serum intervention group was higher than that in the model serum intervention group(P <0.05).Conclusion:1?The expression of Act A increases when Act A/Smads pathway is activated again.Acta is produced by ischemic neurons whicn in turn reacts on neurons themselves and complete the signal transduction from the extracellular cell through transmembrane signal receptor ActRIIA and cytoplasmic Smad3.Signal transduction of Act A/Smads pathway mediated by Act A-mediated neurons in ischemic lesions is target.2?Activation of Act A / Smads signaling pathways that protect neurons by means of cerebral ischemic injury may be achieved by inhibiting the apoptosis of neuronal cells.Increased expression of transmembrane signaling receptor ActRIIA is likely to be the underlying cause of activation of Act A/Smads pathway by cerebral ischemic injury.3?In this experiment,a variety of test results suggest that acupuncture well points can significantly improve the learning ability and memory ability of vascular dementia rats,and have a significant effect on the secretion of Act A from neuronal cells in vascular dementia rats.It stimulates neuronal cells to increase the expression of Act A,which positively regulates the activation level of Act A/Smads signal transduction pathway in order to achieve the role of protective neuronal cells,and ultimately achieve the purpose of treatment of vascular dementia.
