Objective Comparison of single neural stem cells(NSCs)transplantation in the rat spinal cord injury(SCI)and bone marrow mesenchymal stem cells(BMSCs)and between NSCs sequential treatment the curative effect of SCI in rats,To explore the effect of BMSCs microenvironment of spinal cord and the feasibility of sequential treatment.Methods Separation,culture in vitro,purified NSCs and BMSCs,improved Allen method of manufacturing the rat model of spinal cord injury(SCI),48 rats were randomly divided into separate tag neural stem cell transplantation group(group A,n =24),mesenchymal stem cells + tag neural stem cell transplantation group(group B,n =24).Rats of group A in 3 d after spinal cord injury by tail vein injection of NSCs conditioned medium after transfected of slow virus 2 ml,Rats of group B in 1 d after spinal cord injuryby tail vein injection of BMSCs conditioned medium and in 3 d after spinal cord injury by tail vein injection of NSCs conditioned medium after transfected of slow virus 2 ml.By two unwitting graduate to BBB score of two groups of rats,record the data.Respectively in the day before transplantation,three days after transplantation,one week,two weeks,three weeks,four weeks,6 weeks,8 weeks in spinal cord tissue,observation of syringomyelia formation,neural stem cells divide and grow,and spinal cord glial scar size.Results Sequential transplantation group of BBB score higher than the single neural stem cell transplantation group(P < 0.05).Observe the rats in three days after treatment,the blood vessel growth situation when 1,2,3,4,6,8 weeks,spinal cord cavity size A,there are significant differences between group B(P<0.05).Conclusion BMSCs can promote the effective of NSCs differentiation,reduce the tendency of glue,improved the microenvironment of the spinal cord is advantageous to the treatment of SCI,between bone marrow mesenchymal stem cells and neural stem cells sequential transplantation in rat spinal cord injury curative effect is better than that of single neural stem cells transplantation.
