Background and Objective:Psoriasis is a long,stubborn and recurrent polygenic hereditary skin disease that seriously affects the patient’s skin appearance and quality of life,and giving patients a tremendous burden and pressure on the economy and life.The pathogenesis of psoriasis is generally thought to be related to genetic,immunologic,infectious,endocrine,metabolism,environmental,mental psychological and other factors.It is a chronic inflammatory skin disease,duing to hereditary and abnormal immunological factor.Eventually,these factors lead to abnormal proliferation and differentiation of keratinocytes,dermal papillary vascular abnormalities,cuticle parakeratosis and hyperkeratosis,formed a typical clinical psoriasis lesions.The interaction of each factor in which genetic factors are increasingly concerned by domestic and foreign scholars.Recently,with the learning of cell communication study into the people’s vision,some scholars believe that the pathogenesis of psoriasis is relevant with the abnormal inter-cellular communication.And the gap junction is the fundamental structure for inter-cellular communication.Gap junction is a special membrane structure to connect the two adjacent cells,and can be a significant role in the communication and transport of information,energy and substance.And realize the regulating of cell metabolism,homeostasis,proliferation and differentiation.As the basic functional unit,the abnormal of connexin will directly affect the gap junction and the function of GJIC,and resulting in the disturbance of metabolism,homeostasis,proliferation and differentiation.Abnormal proliferation and differentiation of keratinocytes have been found in psoriasis vulgaris.The aim of this study was to investigate the actual situation of Cx protein and its corresponding m RNA in the psoriasis vulgaris lesion,and to find their potential correlation between Cx protein and the severity of disease.Materials and methods:Comparing the expression of Cx26,Cx30,Cx31,Cx43 and their corresponding m RNA between the psoriasis vulgaris group and normal group by the detection by immunohistochemistry and in situ hybridization of nucleic acids through case-control study.Two independent samples of t-test analysis were performed by SPSS 20.0 software,to find the potential significant relevance between two groups.On the basis of the significant differences between the two groups,the PASS score was used to determine the correlation between the number of Cx expression in the experimental group and the PASI score of the psoriasis vulgaris.Results:1.In the immunohistochemistry of skin tissue,Cx26,Cx43,Cx30 and Cx31 were expressed in the epidermis of the skin tissue from the psoriasis group and the normal control group,especially in the spinous layer.These connexins were expressed on the cell membrane,but the strength of expression is different.Cx30 and Cx43 were positively expressed in the skin epidermis of psoriasis vulgaris group and weakly in the normal control group,which was statistically significant.Cx30 and Cx31 were weakly expressed in the skin tissues of the psoriasis group and the normal control group(especially the spine cell layer),and there was no statistical difference.2.In situ hybridization of nucleic acids showed that GJB2,GJA1,GJB6 and GJB3 m RNA were expressed in the epidermis of the skin tissue from the psoriasis group and the normal control group,respectively,especially in the spinous layer.These m RNAs were expressed on the cytoplasm and partial nuclei with different expression strength and weaknesses.Cx26 and Cx43 respectively showed positive expression in the skin epidermis of psoriasis vulgaris group and weakly positive expression in the epidermis of the normal control group,and thedifference was statistically significant.Cx30 and Cx31 respectively showed weakly positive expression in the skin tissues of the psoriasis group and the normal control group(especially the spinous cell layer),and there was no statistical difference.3.There was a postive linear regression relationship between the mean optical density of Cx26 and Cx43 exressed in the skin tissue of the experimental group and the PASI score of psoriasis vulgaris.The regression coefficient between the number of Cx26 expression(mean optical density)and the severity of psoriasis vulgaris(PASI score)was 0.007 and the intercept was 0.344.and the regression coefficient was tested using the t test,t=4.645,p<0.001,which is statistically significant.The regression coefficient between the mean optical density of Cx43 expression and the severity of psoriasis vulgaris(PASI score)was 0.011,the intercept was 0.326,and the regression coefficient was tested using the t test,t=6.000,p<0.001,which is statistically significant.Conclusion:1.Compared with the normal control group,Cx26,Cx43 and their corresponding m RNAs were up-regulated in the lesions of psoriasis vulgaris and the expression of Cx26 and Cx43 were positively correlated with the severity of psoriasis vulgaris.That is to say,the expression of Cx26 and Cx43 were increased as the psoriasis vulgaris grew worse.which can be considered Cx26 and Cx43 may be involved in the pathogenesis of psoriasis vulgaris.2.The expression of Cx30,Cx31 and their corresponding m RNA showed no difference between the psoriasis vulgaris group and the normal control group.There is no reliable evidence of supporting that Cx30,Cx31 and its corresponding m RNA is correlated with the onset of psoriasis vulgaris.
