Objective Coxsackievirus A group 16 virus was cultured by cell culture and identified by RT-PCR,and using the virus to establish a model of mouse viral myocarditis.Study on the Cardiac Tissue and Serology of Mice with Viral Myocarditis:1.The relationship between pathology itself and the immune environment of the disorder and viral myocarditis;2.The appropriate dose of ribavirin in the treatment of mice viral myocarditis infected with CVA16.Methods 1.Through the cell culture technology,culturing the Vero cell which is sensitive to CVA16,and then infected with it.Observing the virus' s pathogenic ability,At the same time,the virus was identified by RT-PCR,and the virus was cultured to the 9th generation.The virus supernatant was centrifuged and sterilized.The TCID50 test was used to detect the virus concentration.2.The mice were infected by intraperitoneal injection using the virus obtained above,and establish an animal model of viral myocarditis.The rats in the experimental group were divided into five groups: experimental group(group A),control group(group B),drug intervention group(group C),middle group(D group)and low dose group(group E).The mice were sacrificed on the 3rd,7th,11 th and 15 th day after birth.The serum and heart tissues were obtained.HE staining was performed on the part of the heart tissue.Another part of the cardiac tissue was used to extract RNA and performed RT-PCR experiment for identification.Serum samples were used to detect IL-6 values in serum using the ELASA kit.Finally using the variance analysis of random block design data and the variance analysis of completely random design data for statistical analysis.Results 1.The virus was identified as CVA16 by RT-PCR and the concentration of the virus was calculated by TCID50 was 106 TCID50 / ml.2.The weight of the mice increased with the age,the control group mice weight and the appropriate dose of ribavirin after intervention than the experimental group of mice weight gain more obvious,and high dose and low dose after intervention with the experimental group mice did not Significant improvement.3.The serum IL-6 level of the mice was first raised,day 7 peak,then begin to decrease,each group was significantly higher than that in the normal group.4.Infection of CVA16 mice in the pathological sections of myocardial tissue can be seen in significant inflammatory lesions,The appropriate dose of ribavirin intervention in myocardial inflammation was significantly improved,After high-dose and low-dose ribavirin intervention,the inflammation was not significantly improved compared with the experimental group.5.RNA was randomly extracted from the myocardium of the experimental group by RT-PCR to obtain the same size band as CVA16.Conclusion 1.Coxsackie virus A group of 16 can be pathogenic by the occurrence of viral myocarditis,At the same time immune within the environment also play a role in myocardial injury;2.The appropriate dose of ribavirin in the treatment of CVA16-related myocarditis has a certain clinical significance.
