| Objective The expression levels of NLRP3 and AIM2 in peripheral blood mononuclear cell(PBMC)and polymorphonuclear(PMN)are compared in patients between rheumatoid arthritis(RA)and normal control group,among rheumatoid arthritis patients containing active group and stable group.To study the pathogenesis of RA in these two inflammatory systems,this article analyzes the correlation between these two inflammatory mediators with laboratory clinical indicators in RA.Methods 49 cases of RA patients were collected from Anhui Provincial Hospital,and the diagnosis was in line with the RA diagnostic criteria of the American College of Rheumatology(ACR)in 1987 and 23 cases of physical examination in the same period from our hospital.According to the DAS28 score,RA patients were divided into two groups: active group of 32 cases and stable group of 17 cases.The clinical data were collected and PBMC and PMN were separated by density gradient centrifugation..The m RNA expression levels of NLRP3 and AIM2 in PBMC and neutrophils of RA and normal controls were detected by RT-PCR.The relationship between NLRP3 and AIM2 was correlated with laboratory clinical data.Results(1)The expression of NLRP3 and AIM2 m RNA in PBMC and neutrophils of RA patients was significantly correlated with the co-signal-related protein containing a CARD(ASC),caspase-1(P <0,05).The expression of Caspase-1 m RNA was correlated(P <0.05).(2)The expression of NLRP3,AIM2,ASC and Caspase-1 m RNA in PBMC of RA patients was significantly higher than that of normal control group,the difference was statistically significant.(0.93(0.78,1.13)vs 0.58(0.54,0.70),p = 0.003;0.81(0.62,1.22)vs 0.60(0.57,0.76),p = 0.012;0.95(0.81,1.28)vs 0.62(0.54(0.65),p = 0.000;0.86(0.79,1.13)vs 0.65(0.55,0.72),p = 0.004).(3)The expression levels of NLRP3,AIM2,ASC and Caspase-1 m RNA in PMN were significantly higher than those in normal control group.(0.97(0.81,1.34)vs 0.61(0.51,0.77),p = 0.005;0.95(0.84,1.25)vs 0.60(0.51,0.71),p = 0.000;0.95(0.82,1.24)vs 0.55(0.48,0.71),P = 0.021;0.93(0.84,1.15)vs 0.62(0.46,0.71),p = 0.001).(4)The levels of NLRP3,AIM2,ASC and Caspase-1 in PBMC of RA patients were significantly higher than those in stable group,the difference was statistically significant.(1.07(0.86,1.26)vs 0.77(0.68,0.84),p = 0.000;1.03(0.73,1.31)vs 0.65(0.56,0.79),p = 0.004;1.10(0.87,1.38)vs 0.83(0.75,,P = 0.008;1.06(0.84,1.35)vs 0.81(0.73,0.88),p = 0.000).(5)The levels of NLRP3,AIM2,ASC and Caspase-1 in PMN were significantly higher than those in stable group,the difference was statistically significant.(1.08(0.86,1.44)vs 0.89(0.80,1.14),p = 0.002;1.10(0.93,1.36)vs 0.85(0.75,0.96),p = 0.000;1.06(0.89,1.28)vs 0.95(0.75,0.98),P = 0.014;1.02(0.87,1.18)vs 0.75(0.71,0.95),p = 0.000)(P <0.05).(6)NLRP3,AIM2,ASC,Caspase-1mRNA in PBMC of RA patients were positively correlated with ESR,CRP,and DAS28(P < 0.05).There was no correlation with anti CCP antibody,rheumatoid factor and course of disease(P > 0.05).(7)NLRP3,AIM2,ASC,Caspase-1mRNA in PMN of RA patients were positively correlated with ESR,CRP,and DAS28(P < 0.05).There was no correlation with anti CCP antibody,rheumatoid factor and course of disease(P > 0.05).Conclusions(1)NLRP3 and AIM2 in PBMCs and neutrophils of RA patients were up-regulated,and the expression of NLRP3 and AIM2 m RNA was correlated with ASC and Caspase-1 m RNA downstream of RA,Neutrophils,NLRP3 and AIM2 may promote the occurrence and development of RA by activating downstream ASC,Caspase-1 and overexpression.(2)The expression levels of NLRP3 and AIM2-m RNA in peripheral blood PBMC and neutrophils of RA patients were positively correlated with ESR,CRP and DAS28,and the levels of NLRP3,AIM2,ASC and Caspase-1 in PBMC and neutrophils of RA patients were significantly higher than those stable group,suggesting that NLRP3 and AIM2 inflammatory body expression levels and RA severity of the disease is to determine the prognosis of the important factors. |
