Objective:This study aims to investigate the effect of silencing PARP1 gene on inhibits proliferation and migaration of gastric carcinoma cell lines SGC-7901 by ?-lapachone.Methods:Using RNA interference technology to transfect the PARP-1-specific small interfeing RNA sequence into gastric carcinoma cell lines SGC-7901 in order to silence the PARP-1 gene expression.Western blot was used to detect the protein expression level of PARP-1 cleavage protein(P89).The gene sequence of the highest transfection efficiency was screened to make the PARP-1 gene silenced MTT assay was used to detect the effect of?-lapachone on the inhibition rate of gastric carcinoma cell lines SGC-7901.Scratch experiment was used to detect the effect of ?-lapachone on the inhibition of migration of gastric cancer cell lines SGC-7901.Plate cloning experiment was used to detect the effect of ?-lapachone on the inhibition cloning ability of gastric carcinoma cell lines SGC-7901.Results:1.Western blot assay provided the expression levels of PARP-1 cleavage of protein(P89).The protein of PARPlsi-RNAl group decreased 33.4%(P<0.05).The protein of PARPlsi-RNA2 group was no signi'ficant difference(P>0.05).The protein of PARPlsi-RNA3 group decreased 50.2%(P<0.01).It is indicating that PARPlsi-RNA3 is the best sequence to silence PARP-1 gene.2.MTT assay showed that silencing PARP1 gene of gastric carcinoma cell lines SGC-7901 could attenuate the cell inhibition rate of ?-lapachone.The most significant difference between experimental group and control group was 4?M drug concentration.The inhibitionrate of the experimental group decreased 32.6%(P<0.01).3.Plate cloning experiment showed that silencing PARP1 gene can could attenuate the inhibition of cloning ability of gastric cancer cell lines SGC-7901 by ?-lapachone.Compared with the experimental group and the control group after 2 weeks,the clone formation rate of the experimental group increased 44.3%(P<0.01).4.Scratxch test showed that the cell migration distance was gradually shortened with time(P<0.05).Scratch experiment showed that silencing PARP1 gene could could attenuate the inhibition of migration of SGC-7901 gastric cancer cell lines by ?-lapachone.The most significant difference between experimental group and control group was the time of 12h(P<0.05).Conclusion:1.Silencing PARP1 gene could attenuate the inhibition of proliferation of gastric cancer cell lines SGC-7901 by(3-lapachone.2.Silencing PARP1 gene could attenuate the inhibition of migration of gastric cancer cell lines SGC-7901 by ?-lapachone.
