Objective:The experiment was to monitor the expression of miRNA-21 and miRNA-451 in serum of esophageal squamous cell carcinoma and to explore the value with diagnosis,clinical pathologic characteristics and the relationship with the efficacy of radiotherapy.Methods: 1.Peripheral blood samples from 50 cases previously untreated esophageal squamous cell carcinoma patients between May 2015 to May2016 from the department of Oncology were collected in our hospital.And 20 age and sex-matched healthy volunteers as controls from medical examination center.2.Real-time quantitative polymerase chain reaction was used to detect the serum level of miRNA-21 and miRNA-451 in esophageal squamous cell carcinomas and healthy controls.The receiver-operating characteristic curves(ROC)are used to calculate the area under the ROC,to diagnostic sensitivity threshold and to determine sensitivity and specificity,and combining with SCC to analysis them comprehensively.3.To monitor the expression of miRNA-21 and miRNA-451 in esophageal squamous cell carcinoma patients after radiotherapy.Results :1.The expression level of miRNA-21,miRNA-451 and SCC in esophageal squamous cell carcinoma(2.33±1.10,3.57±1.98,2.06±2.13)was obviously higher than healthy controls(1.05±0.70,1.05±0.66,0.675±0.36),with statistical significant(P < 0.05).2.MiRNA-21,miRNA-451 and SCC had an area under the curve(AUC)of ROC of 0.845,0.850 and 0.715 with sensitivity of 70%,76% and 55%,and specificity of 85%,85% and 89%,respecitivity.In the combination of miRNA-21,miRNA-451 and SCC had an area under the curve(AUC)of ROC of 0.932,with sensitivity of 80%,and specificity of 90%.3.The expression level of miRNA-21 was significantly associated with clinical stage,tumor differentiation and the lymph node metastasis.And the expression level of miRNA-451 was significantly associated with clinical stage and the lymph node metastasis(P<0.05).4.The expression level of miRNA-21 and miRNA-451 in esophageal squamous cell carcinoma patients after radiotherapy was declined significantly(P<0.05).In the near future curative effect and the expression level of miR-21 and miRNA-451 dynamic changes are consistent.The more effective treatment of esophageal squamous cell carcinoma patients,the more obvious reduction of miRNA-21 and miRNA-451 expression(P<0.05).Conclusion:1.The serum expression of miRNA-21,miRNA-451 and SCC were up-regulated in esophageal squamous cell carcinoma patients compared to healthy volunteers.MiRNA-21 and miRNA-451 can be used as diagnostic biomarkers.2.Up-regulation of serum miRNA-21 and miRNA-451 were associated with some malignant characteristics of esophageal cancer,so they maybe poor prognosis factor in esophageal squamous cell carcinoma.3.MiRNA-21 and miRNA-451 could be useful for evaluating radiotherapy effect in esophageal squamous cell carcinoma.
