Objective:By studying the acute appendicitis bacterial flora distribution and infection of correlational research,to discovery the acute appendicitis bacterial flora distribution,to provide basis the micro ecological prevention and control the acute appendicitis.Methods:Initial screening of the organization of Dalian Municipal Central Hospital 60 cases of appendicitis patients with 3-76 years old from 2016 January to September.The appendicitis patients average age of 15 including the male 37 female 23 people.Then,to extracte total DNA of the appendiceal contents bacteria community,to analyze the bacteria community structural by PCR-DGGE fingerprinting.The appendix organization was fixed,stained with HE staining,then detecting pathological section with light microscope.Results:1.The appendix organization were fixed,stained with HE staining,then detecting pathological section with light microscope into groups of three: 30 cases of acute simple appendicitis;18 cases of acute phlegmonous appendicitis;12 cases of acute gangrenous appendicitis.2.The DGGE fingerprinting showed that the acute simple appendicitis group has the 24.21 ± 3.79 gel strips;and the acute phlegmonous appendicitis group has the15.86±4.56 gel strips;and the acute gangrenous appendicitis group has the 31.75±3.65 gel strips.The analysis results were concluded that the deference of flora diversitydifference of the acute simple appendicitis group and the acute phlegmonous and gangrenous appendicitis group have statistics significance(P<0.05).Phylogenetics is different in the three groups,but not obvious.Each internal flora similarity is greater than the similarity between the sample groups,within the same set of bacterial flora changes there is a certain trend.3.The DGGE fingerprinting showed that the children’s acute appendicitis group has the 24.56±6.10 gel strips;and the adults’ acute appendicitis group has the 18.43± 9.41 gel strips.The analysis results were concluded that the deference of flora diversity difference in both groups have statistics significance(P<0.05).From the age factor,the two groups have different phylogenetics.The flora phylogenetics of adults’ acute appendicitis group is similar.4.The DGGE fingerprinting showed that the male’s acute simple appendicitis group has the 24.31±3.54 gel strips;and the acute phlegmonous appendicitis group has the 16.30±4.96 gel strips;and the male’s acute gangrenous appendicitis group has the 31.90±3.96 gel strips.The analysis results were concluded that the deference of flora diversity difference of the male’s acute simple appendicitis group and the male’s acute phlegmonous and gangrenous appendicitis group have statistics significance(P<0.05).The flora phylogenetics of male’s acute simple appendicitis group compared with others is different.The male’s acute phlegmonous and gangrenous appendicitis two groups have similar flora phylogenetics.5.The DGGE fingerprinting showed that the female’s acute simple appendicitis group has the 24.09±4.06 gel strips;and the female’s acute phlegmonous appendicitis group has the 13.50±3.50 gel strips;and the female’s acute gangrenous appendicitis group has the 31.00 ± 1.00 gel strips.The analysis results were concluded that the deference of flora diversity difference of the female’s acute simple appendicitis group and the female’s acute phlegmonous and gangrenous appendicitis group have statistics significance(P<0.05).The flora phylogenetics of female’s acute simple appendicitis group compared with others is different.The female’s acute phlegmonous and gangrenous appendicitis two groups have similar flora phylogenetics.6.The DGGE fingerprinting showed that male’s a acute simple appendicitis group has the 24.31±3.54 gel strips,and female’s acute simple appendicitis group has the24.09 ± 4.06 gel strips,respectively.The analysis results were concluded that the deference of flora diversity difference of the male’s acute simple appendicitis group and the female’s group have not statistics significance(P>0.05).From the sex factor,the two acute simple appendicitis groups have different phylogenetics.7.The DGGE fingerprinting showed that male’s acute simple appendicitis group has the 24.31±3.54 gel strips;and the male’s acute phlegmonous appendicitis group has the 16.67±4.99 gel strips.The analysis results were concluded that the deference of flora diversity difference of the male’s acute simple and phlegmonous appendicitis groups have statistics significance(P<0.05).The male’s acute simple and phlegmonous appendicitis groups have different phylogenetics.8.The 26 gel strips having obvious specificity and may be dominant bacterial was sequenced.Test results for the 11 gel strips ware doublet.Test results for the remaining15 gel strips having obvious specificity and may be dominant bacterial ware that:Escherichia coli;Coprococcus;Bacteroides;Methylobacterium;Klebsiella;Porphyromonas;Petostreptococcus;Odorous bacillus.The analysis results were concluded that the gram-negative bacteria accounted for 80%,50% of which are Escherichia Coli,the rest20% are gram-positive bacterium.Conclusion:1.The acute appendicitis pathogens is complex.The pathological type of acute appendicitis bacterial flora has obvious change in species and quantity.2.The acute appendicitis bacterial flora diversity associated with the age factor.3.The acute appendicitis bacterial flora diversity associated with the sex factor.4.The acute appendicitis’ pathogenic bacteria is all kinds of gram-negative bacteria of which represented by Escherichia Coli mainly a variety of pathogenic bacteria infection.
