| The main cause of pulpitis is bacterial infection,after the hard tissue of the teeth is destroyed,the bacteria invade the pulp and cause.G-bacteria is the main pathogen of pulpitis,LPS in the cell wall is the important virulence factor.Like other parts of the host,pulp inflammation is the host’s immune response to pathogens.The relationship between destruction and repair of pulp tissue determines the vitality of the pulp tissue and its degree of inflammation.In the host immune response,immune cells and non-immune cells are able to express pattern recognition receptors(PPRs)to identify pathogen and induce inflammatory responses.DC-SIGN is a member of the C-type lectin,which not only activates the initial immune response,but also inhibits the immune response.In recent years,it has received much attention.DC-SIGN is mainly distributed in dendritic cells and macrophages,it has the function of cell adhesion and pattern recognition receptor,and it play a role in the specific and non-specific immune response.At present,the role of DC-SIGN in inflammatory dental pulp is not reported,and its mechanism in the development of dental pulp disease is not clear.This study uses immunohistochemical method to discuss the expression and function of DC-SIGN in the process of human dental pulp disease.Objective:To observed the expression and localization of DC-SIGN in normal and inflammatory human dental pulp by immunohistochemical method.The effect of DC-SIGN on human pulpitis was analyzed statistically.Method:The fresh dental pulp were collected clinically.The histopathological changes were observed by optical microscopy after fixation,embedding,sectioning and Hematoxylin-eosin staining routinely,and the results were taken as the basis for grouping.The specimens were divided into three groups combining with clinical manifestations: the normal pulp group,the acute pulpitis group and the chronic pulpitis,each group contains 20 specimens.Among them,the normal pulp group showed fibroblasts,undifferentiated mesenchymal cells,capillaries and collagen fibers;acute pulpitis group showed vasodilatation congestion,neutrophils,local liquefaction necrosis;chronic pulpitis group showed lymphocytes,plasma cells,macrophages and other chronic inflammatory cells.Immunohistochemical method was used to observe the localization and expression of DC-SIGN in three groups.The photomicrography and Image pro-plus image analysis software was used to evaluated the results.Results:HE staining shows vascular expansion congestion and extensive infiltration of inflammatory cells in acute and chronic pulpitis group,.Immunohistochemical results show that DC-SIGN expression is expressed in the three groups,but the average optical density of different groups was different,DC-SIGN expression in pulpitis is stronger than normal pulp tissue(P<0.05).Conclusions:DC-SIGN is expressed in human inflammatory pulp and play a important role in the development and progression of pulpitis. |
