In recent years,in order to obtain different physiological activity of the steroid compounds,in its parent or branch on the introduction of different functional groups.Steroidal oxime compounds have antibacterial,anti-tumor,anti-virus and other physiological activities.In this study,the steroid oxime derivatives were obtained by biotransformation,which provided material basis and theoretical basis for the research and development of steroidal drugs,and had certain practical application value.A strain capable of transforming steroidal oxime was screened from the soil,which was identified as Penicillium camembert.The investigations were undertaken of the microbial transformation of synthesized 3?-hydroxy-5-ene steroidal oximes with Penicillium camemberts,and the biotransformation products were separated on silica gel column chromatography,the characterizations of the metabolites were identified by various spectroscopic methods.MTT assay was used to detect the inhibitory effect of steroidal ketone raw materials,steroid oxime and biotransformation products on the proliferation of cervical cancer He La cells and human malignant embryonic rhabdomyoma RD cells.3?-Hydroxy-5-ene steroidal oximes were transformed with Penicillium camembert were carried out,in which the steroidal oximes mainly took place 7?-hydroxylation and hydrolysis of hydroxyimino group.A new stable 5-membered heterocyclic compound(9)was obtained from the dehydration of hydroxyimino group with the hydroxyl group which resulted from hydrolization of 16?,7?-epoxy in the biotransformation of 16?,17?-epoxy-progesterone oxime(5).7?,11?-Dihydroxylation and hydrolyzation occurred in the transformation of pregnenolone oxime(6).Dehydroepoxyne oxime(7)mainly took place 7?-hydroxylation and 6?-hydroxylation,and the C-3 hydroxyl group is oxidized to carbonyl group.The results of MTT assay indicated that the steroids displayed time-and dose-dependent inhibitive effect on He La and RD cells.The pregnenolone oxime(6)showed the highest cytotoxicity against the two cell lines,and the IC50 were14.0 ?g/m L and 11.7 ?g/m L,respectively.The 7?-hydroxy-dehydroepoxyne oxime(13)in biotransformation products had the highest cytotoxicity against the two cell lines,and the IC50 were 38.2 ?g/m L and 22.7 ?g/m L,respectively.In summary,a strain capable of transforming steroidal oxime was screened from the soil,and the steroid oxime was biotransformed to obtain a steroidal oxime derivative.The results of in vitro activity test showed that steroidal ketone,steroid oxime and transformation products had certain inhibitory effect on He La and RD cells.
