Atherosclerosis is a common metabolic disorder,which is often associated with diseases such as coronary heart disease,cerebral thrombosis and cerebral ischemic stroke.It could endanger to life when it is serious.Apolipoprotein C?(apo C-?),exists on the surface of low density lipoprotein and very low density lipoprotein,is a small protein whose development is closely related with the incidence of atherosclerosis generally.Tyloxapol is a kind of surface active agent.It can inhibit the activity of enzyme lipoprotein to effect the removal of LDL leading to its accumulation in the blood.Chinese medicine is widely researched and applied in clinical treatment because of the little side-effects and low cost.In this experiment,we used apo C-?-induced vitro model in mice 3T3-L1 cells to observe the release and the m RNA expression of inflammatory factors like TNF-?,IL-1? and IL-6.Then we applied western blot todetect the activity of Nrf2/HO-1 signaling pathways to determine the function and molecular mechanism of aucubin on the mice model.Our results showed that the levels and m RNA expression of TNF-?,IL-1? and IL-6 were significantly increased by apoc-? in the model groups.After the application of aucubin,the levels and m RNA expression were obviously declined compared with the model groups.Results of Western blot showed that the aucubin activated the Nrf2/HO-1 signaling pathways.In addition,in Triton WR 1339-induced mice model,aucubin significantly inhibited the increase of cholesterol(TC),triglyceride(TG),low density lipoprotein(LDL-C)content in the blood caused by Tyloxapol,while improving the level of high-density lipoprotein(HDL-C).By influencing the contents of monocyte chemotactic protein 1(MCP-1),myeloperoxidase(MPO)and superoxide dismutase(SOD)enzyme activity,aucubin improved the resistance to oxidative stress and anti-inflammatory abilities of body.Aucubin also reduced the purity of apo C-?and liver index.Results of frozen section revealed the aucubin could significantly reduce the level of liver lipid.To sum up,aucubin performed the function of anti atherosclerosis by its obvious anti-inflammation and antioxidant activity,which may become a kind of new drug targeting to atherosclerosis.
