Objective: To explore the effect of Bone Marrow Mesenchy malstem Cells(BMSCs)with ApoJ gene in experimental intracerebral hemorrhage(ICH)in rats.Methods: The bone marrow mesenchymal stem cells(BMSCs)were cultured in vitro,recombinant plasmid pEGFP-N1-apoJ was transferred into BMSCs with lipofectamine in vitro,transfection efficiency of different recombinant plasmids with the same volume liposome were observed under the fluorescence microscope at 24 h,48h,72 h and 96 h,the transfection group that transfection rate was highest was screened out.102 male SD rats were randomly divided into three groups: transfection group,BMSCs group and NaCl group.Each group was divided into 24 h,48h,72 h and 96 h four subgroups.Rats ICH models were established through brain stereotactic instrument.The three groups were injected with the same volume of transfected cell suspension,BMSCs suspension and normal saline in 24 hours after modeling by intracranial injection,and then these rats of each corresponding subgroups were sacrificed at the end of intervention 24 h,48h,72 h and 96 h.Modified neurological function score(mNSS)was performed at each time point;dry and wet quality method was employed to measure brain water content and expression level of ApoJ at each time point weredetected by Western blot.Results: 1.Under the condition of equal volume liposome,transfection efficiency of the four transfection group(BMSCs were transfected with recombinant plasmid were 0.8,1.2,1.6,2.0)in the 72 h was higher than the same group in 24 h,48h,96 h,and compared with the other three cell transfection groups(BMSCs were transfected with recombinant plasmid were 0.8,1.2,,2.0),The fluorescence was the strongest of the transfection group with recombinant plasmid was 1.6,and the transfection efficiency was higher than other three groups at the same time points(P < 0.05);2.mNSS in transfected group were significantly reduced compared to BMSCs group and the NaCl group at the same time point at days1,3,5 and 7 after the experimental intervention(P < 0.05);3.The results of brain tissue water content showed that the transfected group were significantly reduced compared to BMSCs group and the NaCl group at the same time point at days1,3,5 and 7(P<0.05);4.The results of Western blot showed that the expression level of ApoJ in the brain tissue of the transfected group was higher than that of the BMSCs group and the NaCl group at the same time point(P < 0.05).Conclusion: Bone Marrow Mesenchy malstem Cells with ApoJ gene were able to expression apolipoprotein J in brain tissue of ICH rats.Exogenous ApoJ has a therapeutic effect on ICH,which can promote the recovery of neurological function after ICH in rats,relieve cerebral edema,and improve the prognosis of ICH.
