Objective: The analysis of the effect of Shugan Bushen recipe on embryo implantation dysfunction induced by psychological stress and its influence of CD4~+Foxp3~+Treg cell,which is based on Xiaoyao Powder and Shoutai pill,to explore study on the effect of Shugan Bushen recipe on improving the embryo implantation dysfunction induced by psychological stress.Methods: In the experiment,the rats were treated with allogeneic mating(BALB/c×C57BL/6).The SPF level of female BALB/c rats,two months old.The SPF level of male C57BL/6 male rats,three months old,mating with BALB/c female rats.The SD male rats in the SPF grade,mass 300 g,used to cause psychological stress in rats.The normal control group,the psychological stress group,the psychological stress+Shugan recipe group,the psychological stress+Bushen recipe group,the psychological stress+Shugan and Bushen recipe group,24 rats in each group.Female rats were anesthetized by intraperitoneal injection of 1% pentobarbital sodium,injection dosage of 0.06g/kg.The iliac lymph nodes and spleen of 12 rats were separated.which used to detect the ratio of Treg cells by flow cytometry.Results:(1)The effect of Shugan Bushen recipe on implantation in pregnant rats:Compared with the normal control group,the implantation site number and the number of implantation of the embryo of the psychological stress group after 5.5 days and 8.5 days of pregnancy were significantly increased.The difference was statistically significant(P<0.05).Compared with the psychological stress group,the implantation site number and the number of implantation of the embryo of the psychological stress+Shugan recipe group and the psychological stress+Bushen recipe group after 5.5 days and 8.5 days of pregnancy were significantly elevated(P<0.05).Compared with the psychological stress+Shugan recipe group and the psychological stress+Bushen recipe group,the implantation site number and the number of implantation of the embryo of the psychological stress+Shugan Bushen recipe group after 5.5 days and 8.5 days of pregnancy were also enhanced obviously.The difference was statistically significant(P<0.05).However,the implantation site number and the number of implantation of the embryo of the psychological stress+Shugan recipe and Bushen recipe group after 5.5days and 8.5 days of pregnancy were lower than the control group.The difference had statistical significance(P<0.05).Compared with the psychological stress+Shugan recipe group,the implantation site number and the number of implantation of the embryo of the psychological stress+Bushen recipe group after 5.5 days and 8.5 days of pregnancy did not change markedly.The difference was not statistically significant(P>0.05).(2)The effect of Shugan Bushen recipe on the proportion of CD4~+Foxp3~+Treg cells in different parts of rats in each group after 5.5 days of pregnancy:Compared with the normal control group,the proportion of CD4~+Foxp3~+Treg cells in spleen,iliac lymph nodes and of the psychological stress group rats after 5.5 days of pregnancy were significantly increased.The difference was statistically significant(P<0.05).Compared with the psychological stress group,the proportion of CD4~+Foxp3~+Treg cells in spleen,iliac lymph nodes and of the psychological stress+Shugan recipe group and the psychological stress+Bushen recipe group rats after 5.5 days of pregnancy were significantly increased(P<0.05).Compared with the psychological stress+Shugan recipe group and the psychological stress+Bushen recipe group,the proportion of CD4~+Foxp3~+Treg cells in spleen,iliac lymph nodes and of the rats after 5.5 days of pregnancy were Significantly increased.The difference was statistically significant.Nevertheless,the proportion of CD4~+Foxp3~+Treg cells in spleen,iliac lymph nodes and of the psychological stress+Shugan Bushen recipe group rats after 5.5 days of pregnancy were lower than the normal control group.The difference was statistically significant(P<0.05).Compared with the psychological stress+Shugan recipe group,the proportion of the cells of CD4~+Foxp3~+Treg in spleen,iliac lymph nodes and of the psychological stress+Bushen recipe group rats after 5.5 days of pregnancy had no marked change.The difference was not statistically significant(P>0.05).Conclusion:Psychological stress leads to decline in the implantation site number and the number of implantation of the embryo of the psychological stress group after 5.5days and 8.5 days of pregnancy.It can also reduce the proportion of CD4~+Foxp3~+Treg cells in spleen,iliac lymph nodes and of the rats after 5.5 days of pregnancy.However,combined use of Shugan Bushen recipe can increase the implantation site number and the number of implantation of the embryo of the psychological stress group after 5.5days and 8.5 days of pregnancy.It can also increase the proportion of CD4~+Foxp3~+Treg cells in spleen,iliac lymph nodes and of the rats after 5.5 days and 8.5 days of pregnancy.Shugan Bushen recipe can promote the embryo implantation by increasing the proportion of CD4~+Foxp3~+Treg cells in pregnant mice.
