Objective: The study was designed to explore the effects of elastin peptides on the expression of costimulatory molecules on murine bone marrow dendritic cells (mDCs) in vitro.Methods: Murine bone marrow dendritic cells were cultured for 7 days in RPMI1640 medium with 40ng/mL GM-CSF, lOng/mL IL-4 and 10% fetal calf serum(FBS). In day 7, High purity of immature DCs coincided with experimental demands were randomly divided into control group and elastin peptide group. Elastin peptide group was stimulated with lug/mL elastin peptides (VGVAPG). DCs of the two groups were cultured for further 24 hours under the same condition. In day 8, DCs of both groups were collected and the expression of costimulatory molecules including CD40, CD80 and CD86 on DCs were detected by flow cytometry and real-time PCR (RT-PCR). The levels of interleukin-6(IL-6) and transforming growth factor ? (TGF-?) were examined by enzyme-linked immunosorbent assay (ELISA).Results: Compared with control group, the levels of costimulatory molecules including CD40, CD80 and CD86 on DCs in elastin peptide group increased obviously (p values all <0.05) and a similar increase in the levels of IL-6 and TGF-P was observed in elastin peptide group (p values all<0.05).Conclusions: (1) The results impaly that elastin peptides can increase the expression of costimulatory molecules on mDCs, including CD40, CD80 and CD86; (2) Elastin peptides can contribute to the secretion of inflammatory cytokines IL-6 and TGF-p of mDCs.
