Objective: To vestigate the effects of Chaihushugansan on apoptosis of gastric smooth muscle cell(SMC)and expression of NF-κB protein in rats with functional dyspepsia.Methods:Totally 48 healthy SD rats were randomly divided into the normal group with 8 rats each,the control group,the domperidone group,the low/medium /high dose group of Chaihushugansan.The FD models were made by clasping their tails.The other FD rats were treated with domperidone suspension and Chaihushugansan decoction were administered respectively to five groups by gavage.Model in 4 weeks,HE staining in gastric tissue pathological change.Transmission electron microscopy(TEM)was employed to observe the mitochondria of the rats’ gastric smooth muscle cells form.TUNEL staining was employed to detect the apoptosis ofsmooth muscle cells.Immunnohistochemistry was used to detect Bcl-2,Bax and Nf-kb expression.Results:Pathological observation of gastric tissue in each group showed no erosion,ulcer,metaplasia and other organic pathological changes.The observation of the gastric tissue showed that the gap between the gastric SMC of the normal group was normal,the connection was close and the mitochondrial morphology was normal by transmission electron microscope.In the model group,the SMC space was widened,the connection was loose,the mitochondria were obviously swollen,and the apoptotic bodies could also be seen.Chaihushugansan groups and the domperidone group all the gastric SMC space were normal,the connection were close,the swelling of mitochondria decreased and the apoptotic bodies decreased.Compared with the normal group,the gastric emptying were lower(P <0.05)in model group.The mitochondria of gastric smooth muscle cell Serious injury,and the apoptosis rate of gastric smooth muscle cell was increased(P<0.05)in model group.The expression of Bcl-2 proteins was decreased(P<0.05),and the expression of Bax proteins was increased(P<0.05).Compared with the model group,Chaihushugansan groups and the domperidone group all the gastric emptying were increased(all P <0.05),and some mitochondria of the gastric smooth muscle cell were significantly improved.The apoptosis rate of gastric smooth muscle cell was decreased(P<0.05),the expression of Bax proteins was decreased(P<0.05),but the expression of Bcl-2 protein was increased(P<0.05).Compared with domperidone group,the gastric emptying and the expression of Bcl-2 proteinwas increased in Chaihushugansan high-dose group(P<0.05),but the apoptosis rate of gastric smooth muscle cell and the expression of Bax proteins was decreased(P<0.05).Compared with the Chaihushugansan low-dose group,the gastric emptying and the expression of Bcl-2 protein was increased in Chaihushugansan high-dose group and medium-dose group(P<0.05),but the apoptosis rate of gastric SMC and the expression of Bax proteins was decreased(P<0.05).Comparison between medium and high dose group are not statistically significant.Conclusion : 1.Chaihushugan play a role in promoting gastric motility by reduceing the apoptosis rate of gastric smooth muscle cells in FD rats,which is to regulate the apoptosis of mitochondrial pathway by up regulating the expression of Bcl-2 and inhibiting the expression of Bax.2.Chaihushugan protect the gastric smooth muscle of FD rats to promote the recovery of gastrointestinal motility,which inhibit the increase of inflammatory reaction and the amplification effect of apoptosis signal by reducing the expression of NF-kappa B protein.
