Objective : To explorethe effects of tea polyphenols(TP)and MAPK(ERK1/2,p38 and JNK)signal pathway inhibitors improving the expression of superoxide dimutase and inhibiting cell apoptosis in brain tissue after cardiopulmonary resuscitation(CPR).Methods:Cardiac arrest/cardio pulmonary resuscitation(CA/CPR)model was established using the method of alternating current-esophagus stimulation.Rats were randomly divided into six groups:model group,PD group,SP group,SKF group and TP group.Sham group was treated without CA/CPR(n = 6).Once spontaneous circulation restoring,the rats of the five groups were injected saline(n=24),PD(10mg/kg,n=24),SP(6mg/kg,n=24),SKF(2.5mg/kg,n=24)and TP(10mg/kg,n=24).The cerebral tissue of superoxide dismutasefluorescence density and apoptotic cells were detected with immunofluorescence labeling method and Caspase3/TUNEL double labeling method.Results:1.Compared with CA group,after 12 h,24h,48 h and 72 h with CPR,TP treatment significantly increased the SOD1 fluorescence density;Compared with CA group,after 12 h,24h,48 h and 72 h with CPR TP treatment significantly increased the SOD2 fluorescence density;after72h with CPR the apoptosis rate was significantly decreased.2.Compared with CA group,after 12 h,24h,48 h and 72 h with CPR PD treatment significantly increased the SOD1 fluorescence density;Compared with CA group,after 12 h,24h and 48 h with CPR TP treatment significantly increased the SOD2 fluorescence density,after72 h with CPR the apoptosis rate was significantly decreased.3.Compared with CA group,after 12 h,24h,48 h and 72 h with CPR SKF treatment significantly increased the SOD1 fluorescence density;Compared with CA group,after 12 h,24h,48 h and 72 h with CPR SKF treatment significantly increased the SOD2 fluorescence density,but can not reduce the rate of cell apoptosis compred to CA group after72 h with CPR.4.Compared with CA group,after 12 h,24h,48 h and 72 h with CPR SP treatment significantly increased the SOD1 fluorescence density;Compared with CA group,after 12 h,24h,48 h and 72 h with CPR SP treatment significantly increased the SOD2 fluorescence density,butcan reduce the rate of cell apoptosis compred to CA group after 72 h with CPR.Conclusions:The expression of SOD was decreased and the apoptotic rate was increased in brain tissue after CPR.TP treatment can promote the expression of SOD,thereby decrease the apoptotic rate in brain tissue after 72 h with CPR.ERK signal pathway inhibitor PD98059 treatment can increase the expression of SOD,decreases the apoptotic rate after72 h with CPR.P38 signal pathway inhibitor SKF86002 treatment increase the expression of SOD,but can not reduce the apoptotic rate after 72 h with CPR.JNK signal pathway inhibitor SP600125 treatment decreased the expression of SOD,but can decrease the apoptotic rate after 72 h with CPR.
