The Experimental Study On Mechanical Phlebitis Caused By Mirabilite Externally Applied For The Prevention Of Intravenous Therapy Of Indwelling Needle

ObjectivesBy applying animal model of mechanical phlebitis caused by venous indwelling needle,the thesis mainly observes the vein injury caused by application of mirabilite and the intervention effect of interleukin-1(IL-1),interleukin-6(IL-6),tumor necrosis factor alpha(TNF-α)in vein vascular tissue.Meanwhile,this paper will discuss the prevention and treatment of phlebitis caused by indwelling needle applied by mirabilite in vein,and provide experimental data for the prevention and treatment of venous indwelling needle caused mechanical phlebitis.MethodsBy choosing 57 healthy New Zealand rabbits with big ears,this experiment randomly divides them into 3 groups,one is the control group(9rats),the second is an indwelling needle group(24 rats),and the third is mirabilite group(24 rats).The ear vein of each rabbit will be retained in the indwelling needle group and mirabilite group.0.9% of Sodium Chloride will be injected daily in the indwelling needle group.Fixed external mirabilite application will be given in the mirabilite group at the base of indwelling needle group.On the 3d,5d,7day,three samples will be chosen in the blank control group randomly to draw venous specimen which will be made into histopathologic slide.after ear vein specimens were observed,the expression of IL-1,IL-6,TNF-α will be tested in the rabbit ear vein through immunofluorescence.ResultsThe results of HE staining: no infiltration of inflammatory cells in the blank control group,no hyperplasia of fibrous tissue around the vascular wall.With the increasing of indwelling time,the indwelling needle group and mirabilite group with ear vein increased in fibrous tissue vascular wall,inflammatory cell infiltration in vascular tissues,and organized thrombosis.Compared with treatment group and indwelling needle group,fibrous tissues in the vascular wall are decreased,inflammatory cell infiltration is significantly decreased.The result of Immunofluorescence test: At the same retention time,the TNF-α expression in indwelling needle group and the mirabilite group IL-1,IL-6,are significantly higher than control group(P < 0.05).TNF-α expression in mirabilite group IL-1,IL-6 is lower than that of indwelling needle group(P < 0.05).The IL-1,IL-6,TNF-α expression increase with the growing of the retention time(P < 0.05)in indwelling needle group and mirabilite group.ConclusionsMirabilite can significantly reduce the inflammatory cells infiltration of rabbit ear vein,formation of fibrous tissue hyperplasia and thrombosis of vascular wall.Mirabilite can significantly reduce the expression of IL-1、IL-6 and TNF-α of venous phlebitis caused by indwelling needle mechanical tissue,and reduce the damage degree of rabbit ear vein.
