Objective:To explore the influence of IL-8 and TNF-a in hyperoxia-induced lung injurg in preterm rats.Methods :One-day-old 100 health preterm SD rats were selected. Then its divided randomly into air group and hyperoxia group(both n=50). Rats in the hyperoxia group were exposed to 95% 02, while rats in the air group were raised normally in air. Two group maintinaed the same temperature(26-18?)and humidity(50%-70%), and use soda lime to absorb carbon dioxide,salici gel absorb mositore. Everday, out of the box 1 hour in time to addition the water and feed in time,and change the motherrats to avoid the nuring care deserse. The rats in each group were sacrificed at 1?3?6?10 and 14 days after exposure(10 rats each time point) ,then enchase centrifuge , the liquid supernatant put in the refrigerator(-20?). To using ELISA to check IL-8 and TNF-a.Results:The performance of newborn mice: campare to the normal group,the rats of High oxygen group is good growth situation and activity, and the muzzle is ruddy color,and defecate normal diet. Its no obvious difference with the normal group. When 3 days its began to appear listlessness,decreased activity, diet, breathing becomes shallow, nose and mouth becomes rosy. To 14 days of growth retardation, lethargy, weight does not increase, fast shallow breathing, oral cyanosis, poor diet, defecate dry.The changes of TNF-a and IL-8 in Alveolar lavage fluid: campare to the normal group, high oxygen groups of alveolar lavage IL-8 and the level of TNF-a was high. In the 1 day of high oxygen group, the difference of TNF-a is statistically significant(P<0.05),is to the peak in the 3 day, continued to 6 days, then gradually decline. Nevertheless,the IL-8 of hyperoxia group in 1 day is no statistical significance,and 3 days its has significance(P<0.05),its to the peak when 6 days and continued to 14 days.Conclusion:It is successful of this experiment established high oxygen lung injury animal model, the physiological state of the oxygen with the extension of time,the mice gradually deteriorated.In the early stage of exposuring to hyperoxia, the TNF-a is increased with the extension of exposure time, but its role in the late exposure is weakened, Explain that TNF-ais work in high oxygen lung injury early work.After High oxygen exposure, The concentration of IL-8 is no obvious change in the early stage,the effect is distinct in the late, instructions IL - 8 in lung inflammation of the hair should be continued role.
