ObjectiveEvaluate the expression of PI3Kp 110a and PI3Kp 110β in colorectal serrated lesions, conventional adenoma and adenoma with canceration. Analysis of differential expression between the serrated neoplasia pathway and the conventional adenoma-carcinama sequence, it is to investigate the relationship between PI3Kp110 and the factors of clinical pathology and significance. Analysis its difference among groups, explore the effect of expression of PI3Kp 110α and PI3Kp 110β in the different cancer pathways, it is to provide a theoretical basis for further study of colorectal adenoma carcinogenesis mechanism, and find a reliable marker of adenoma canceration and choose the right treatment options. It has important clinical significance for the prevention of the evolution of colorectal adenoma to adenocarcinoma.Materials and MethodsA total of 520 colorectal tissue samples at different stages were collected from the Department of Pathology of Binzhou Medical University Hospital and Binzhou People’s Hospital between January 2009 and August 2014. Parts of the tissue samples were immediately frozen in liquid nitrogen, and parts were routinely fixed in 10%buffered formalin liquid. Immunohistochemistry and Western blot analysis were conducted to detect the expression of p110α and p110β in normal colorectal tissues,serrated lesions, conventional adenoma and adenoma canceration. Multiple linear regression analysis was used to analyze the relationship between p110α, p110β and the factors of clinical pathology.ResultsResults revealed that the expression of P110α and P110β in the adenoma canceration was significantly higher than that in normal tissues, tubular adenoma (low grade) and tubular-villous adenoma (low grade) of conventional adenoma,hyperplastic polyps of serrated lesions. But there was no significant difference between the adenoma canceration and the high grade of adenoma of conventional adenoma, all grade of villous adenoma and serrated adenoma. Expression of p110αand p110β is relevant to tumor size in all adenoma groups, and relevant to grade and amount of the adenomas in tubular-villous adenoma (TVA). The expression levels of p110α and p110β are high in non-sessile villous adenoma (VA). The expression of p110α and p110β is closely related to differentiation degree and lymph nodular metastasis in cancerated adenoma groups of both pathways.Conclusion1. PI3Kp110α and PI3Kp110β were highly expressed in serrated adenoma and adenoma with canceration of "serrated pathway",suggesting that both play an important role in "serrated pathway" occurrence development.2. The expressions of PI3Kp110α and PI3Kp110β were increased gradually in adenoma-carcinama sequence, its high expression may be one of the factors promoting the dynamic of colorectal cancer beginning.3. ’PI3Kp110α and p110β were highly expressed in serrated adenoma, villous adenoma and adenoma with canceration. Its high expression may be an important cause of what canceration rate of SA and VA was higher than that of other adenomas.4. In the initial stage of the two approaches, PI3Kp110a and PI3Kp110β positive correlation between the expression,there is a synergistic effect in promoting tumor progression.5. Expression of p110α and p110β is relevant to tumor size, grade and number in adenoma , and closely related to differentiation degree and lymph nodular metastasis in cancerated adenoma groups of both pathways.6. PI3Kp110α and PIBKp110β detection can be an important indicator for early warning and prognosis.
