Objective To study on the association between the methylenetetrahydrofolate reductase(MTHFR)C677T gene polymorphism and the concentration of plasma homocysteine(Hcy)with Amnestic mild cognitive impairment(aMCI).Investigate the influence of Hcy and MTHFR C677 T gene polymorphism on the occurrence and development of aMCI to provide evidence for clinical diagnosis and intervention.Method The case-control study was used,65 patients with aMCI as aMCI group,61 patients with AD as AD group,and 63 healthy elderly people were selected as cognitive normal group(control group)the same period.The general clinical information of the observed objects were collected,plasma Hcy was determined by enzyme linked im-munosorbent assay(ELISA),Polymerase chain reaction and restriction fragment length polymorphism(PCR-RFLP)wasused to analyze the MTHFR C677 T genotypes.The Hcy levels,genotypes,allele frequency,and Hcy levels of different genotypes were compared.The regression analysis was used to study the relationship between MTHFR C677 T gene polymorphism and the risk of aMCI.Results When comparing the general data of the three groups,the difference of age,BMI and Hcy was statistically significant(P<0.05);The Pearson correlation analysis showed that the Hcy level was negatively correlated with the MMSE score;The frequencies of TT genotype and T allele of MTHFR gene in aMCI group and AD group were significantly higher than those of control group,the differences were statistically significant(P<0.05).There was difference of the plasma homocysteine levels in observation with TT?CT and CC genotype(P <0.01),there was no significant difference of the plasma homocysteine levels with CC and CT genotype(P >0.05).The relevance of polymorphism of MTHFR C677 T gene and aMCI showed that two kinds of mutation(CT+TT)and the T allele is associated with the risk of aMCI(P < 0.05).Conclusion The plasma Hcy level was negatively correlated with cognitive function,MTHFR C677 T gene mutation in plasma Hcy level are associated with cognitive dysfunction,MTHFR polymorphism of C677 T gene may be an important risk factor for the onset of aMCI.
