Objective:We aimed to analyze the difference of the expression of BRAF gene in bone metastasis and without bone metastasis of prostate cancer by immunohistochemistry and PCR.The correlation between BRAF mutant gene and bone metastasis of prostate cancer was analyzed.Methods(1)A total of 103 patients with prostate cancer who were diagnosed by prostate biopsy or postoperative pathology were enrolled in this study.Twenty patients with benign prostatic hyperplasia were randomly selected from the first affiliated hospital of Nanchang University in January 2013 to February 2015.The bone metastases was diagnosed by ECT,CT / MRI or bone biopsy.Clinical stage of prostate cancer was scheduled according to the American Cancer Society(AJCC,2009)TNM staging criteria.Before surgery,all cases did not receive all types of chemotherapy,radiotherapy and other related anti-tumor therapy.(2)Immunohistochemical method was used to detect the expression of BRAFprotein in bone metastasis,prostate cancer without bone metastasis and benign prostatic hyperplasia(prostate tumor tissue),and to analyze its significance.(3)The mutation of BRAF gene in bone metastasis,prostate cancer without bone metastasis and benign prostatic hyperplasia(prostate tumor tissue)was detected via PCR,and to analyze its significance.(4)Using SPSS 19.0 to establish a database and analyze the data statistically.To study the relationship between BRAF mutant gene and bone metastasis of prostate cancer.Result:1,immunohistochemical results show that:(1)The expression of BRAF protein in prostate cancer with bone metastases and prostate cancer without bone metastases showed that BRAF was statistically different(P <0.05).(2)The expression of BRAF protein in bone metastases of prostate cancer and benign prostatic hyperplasia showed that it was statistically significant(P <0.05).2,PCR results showed that :There did not find BRAF gene mutation in benign prostatic hyperplasia group.BRAF gene mutation in prostate cancer bone metastases group and prostate cancer without bone metastasis group showed that BRAF gene mutation was statistically significant(P <0.05).Conclusion:The expression of BRAF protein in prostate cancer was higher than that in prostate cancer without bone metastasis and benign prostatic hyperplasia.And BRAF gene mutation in prostate cancer was significantly higher than that in prostate cancer without bone metastasis which is suggesting that BRAF gene may be a related factor in prostate cancer patients with bone metastases.
