Objective:There are two objectives in this research,one is to observe the coagulation function of sepsis which could be deduct from measuring TNF-?,P-selectin,TAT,PT,APTT,PLT(We used improved CLP to produce sepsis rats)from rats groups in our test,another objective is to test how ulinastatin affect Coagulation function for sepsis rats which is tested out by applying different dose of ulinastatin to our sepsis rat group to have comparison.Methods:1.Grouping: 120 four-week-old healthy clean grade male rats were selected and divided randomly and evenly into 5 groups: blank control group(A group),sham-operation group(B group),sepsis group(C group),low dose ulinastain intervention group(D group)and high dose ulinastain intervention group(E group),all of these rats are weighed about 120+-20 g.At different time points(6 h,12 h and 24 h)8 rats were randomly selected from heart blood,then the samples were taken to inspect in order.2.Observation index:measure TNF-?,P-selectin,TAT,PT,APTT,PLT for those blood samples,utilize statistical analysis method to visualize the difference between our 5 groups rats.Results:1.Improved CLP do works super well for producing sepsis rats.2.TNF-? for B group is higher than A group,this is no sense.C group has increasing TNF-a when in 6 hour,and continue increasing as time going,reach a peak at 24-hour interval.D group also has a increasing TNF-? when in 6-hour interval,peaked in 12-hour interval,but decrease in 24-hour interval.E group has the same trend as D group,but TNF-a are has a lower value for all time interval(P<0.05).3.There is no point to measure or compare P-selectin for A group and B group.On the contrary,P-selectin is a important index for other groups.C group has radical increase at 6-hour interval,peaked at 12-hour interval,but dropped back at 24-hour interval.D group exposes the same trend as C group,but all values are smaller when compared at the same time point(P<0.05).E group again the same trend,but corresponding value is future smaller,there is really statistic difference between these different values.4.TATs for A group and B group are almost the same,C group has increased TAT at 6-hour interval(P<0.05),peaked at 24-hour interval,and there is a really big different between 6-hour interval and 12/24-hour interval(P<0.05).D group also has an increase at 6-hour interval,but the value is quantitatively smaller than the one in C group,12-hour and 24-hour interval,the trends are similar to C group,but again,values are quantitatively smaller than corresponding ones.E group has the same trend as D group,but values are again quantitatively smaller(P<0.05).It has statistical difference?5.PT/APTT,PLT are almost the same for A group and B group(P>0.05).for C group,when measured in 12 hour interval,PT and APTT significantly increased,PLT decreased(P<0.05),when measured in 24 hour interval,PT continued increased,but APTT showed a decreasing trend,PLT decreased even more;for D group,PT?APTT and PLT all increased both in 12 hour interval and 24 hour interval.Increasing trend was similar to corresponding increasing trend shown in C group,but PT and APTT were quantitatively lower than C group(P<0.05),PLT was quantitatively higher than C group.Analysis of PT,APTT and PLT for E group shown similar trends to D group,but PT and APTT were quantitatively lower(P<0.05),PLT was quantitatively higher(P<0.05),which were really remarkable differences.Conclusions:1.Improved CLP do works super well for producing sepsis rats.2.The function of blood coagulation in sepsis rats is abnormal in 6 hours.3.ulinastatin invention do help preserve Coagulation function for CLP produced sepsis rats.higher dose has more remarkable effect.
