| Non-alcoholic fatty liver disease(NAFLD)is a clinic syndrome characterized excessive lipid accumulation in liver without excessive alcohol or any other identifiable causes,includes non-alcoholic fatty liver(NAFL),non-alcoholic steatohepatitis(NASH),hepatic fibrosis,hepatic cirrhosis and hepatocarcinoma.NAFLD is strongly related to age,obesity and metabolic diseases.Besides controlling body weight and food intake,NAFLD can be improved by insulin sensitizer(like biguanide and thiazolidinedione),hypolipidemic drugs(like fibrates,statins),cytoprotective agents,and antioxidants,etc.In this study,two kinds of NAFL mouse models,including KKAy mice,which induced by high-calorie-diet and characterized with fatty liver,diabetes,obesity,hypertriglyceridemia,and hypercholesterolemia,and Model-T mice,which induced by both high-fat-diet and tetracycline in C57BL/6 mice,were used.The lipid accumulation in liver was evaluated through histological examination(H&E staining),content of hepatic triglyceride,and the echo-intensity attenuation coefficient of liver ultrasound.Serum lipid disorders were estimated by the levels of triglyceride,total cholesterol,high density lipoprotein cholesterol(HDL-C),and low density lipoprotein cholesterol(LDL-C).Glycometabolism was evaluated by fasting blood glucose and glucose tolerance test(GTT).The values of serum insulin and homeostasis model assessment(HOMA-IR)were used to appraise insulin sensitivity,respectively.The results showed RP2014007,the extraction of Rhodiola crenulata,could improve liver lipid accumulation in both NAFL mouse models,KKAy mice and Model-T mice;it also affects hyperglycemia,hypertriglyceridemia,hypercholesterolemia,and insulin resistance in KKAy mice,respectively.Usually,the liver pathologic changes of NASH patients appear lipid accumulation,inflammation,collagen deposition etc.To establish a NASH model,the high-fat-diet and tetracycline were administrated in C57BL/6 mice and KKAy mice,respectively.The hepatic histological examimation(H&E staining)and its semi-quantitative scores of NASH,the levels of serum aminotransferases,total cholesterol,triglyceride,glucose and insulin were measured;and the value of HOMA-IR was calculated.As the results,tetracycline could enhance liver lipid accumulation,and accelerate the progress of NAFLD in both KKAy mice and C57BL/6 mice.But the experimental skills for the utilization of tetracycline,such as the dose,the frequency,should be investigated further,as well as the special technique for evaluating hepatic inflammation. |
