Red blood cells(RBCs),whose can transport oxygen and carbon dioxide,are blood components.Clinically,the need for blood is greater when there is a large amount of blood loss,severe anemia,or in the case of a serious infection.It is very important to storage red blood cells effectively in vitro to ensure using blood.The cryopreservation of RBC is one method of storaging red blood cells.Currently,high concentrations of glycerin are used as the main protectants for frozen red blood cells.High concentrations of glycerol solution of high osmotic pressure and the effect on cell without stability,useing the glycerol cells,cells need to regulate cell osmotic pressure and maintain normal form.Ectoine is one of compatible solutes whose can regulate osmotic pressure in the halophilic bacteria and halotolerant bacteria.According to other studies,we speculate ectoine can regulate osmotic pressure under high concentrations of glycerol.Objective:To investigate the effect of osmotic adjustment of the red blood cells with ectoine and to investigate the quality of red cells when ectoine,as to adjust osmosis pressure,was used to cryopreservate red cells.Methods:RBC kinetics in the isotonic ectoine,in the isotonic sodium lactate,in the nomal saline,in ultrapure water and in the isotonic glycerol were tesed.We ascertained the procedures of glycerolization before freezing RBC and the procedures of deglycerolization after thawing RBC.1.5%,3.0%and 4.5%(are w/v)ectoine with 57%(w/v)glycerol were set as the experimental groups,while the commercial compound glycerin was set as the control group.The recovery rate,the deformability,the osmotic fragility,the content of adenosine triphosphate(ATP),the activity of fructose-6 phosphate kinase(PFK),the activity of pyruvate kinase(PK),and the activity of lactic dehydrogenase(LDH)were tesed at last.Results:The RBC kinetics in the isotonic ectoine,in the nomal saline,and in the isotonic sodium lactate are similar;the RBC kinetics in ultrapure water and in the isotonic glycerol are similar.3%ectoine with glycerol cryopreservated red cells was better than the commercial compound glycerin in the quality of frozen RBC.Conclusion:The isotonic ectoine has the osmotic adjustment function of red cells.3%ectoine with glycerol cryopreservated red cells had improved the quality of the red cells,which can broad the ectoine scope.
