Clinical Observation Of Gemcitabine And Oxaliplatin Combined With Oral Dosage Of Fluorouracil In First-line Chemotherapy Of Pancreatic Cancer

Objective:To observe the efficacy and safety of Gemcitabine and Oxaliplatin combined with oral dosage of Fluorouracil in first-line treatment of pancreatic cancer.Methods:From January 2011 to January 2017,31 patients with locally advanced or metastatic pancreatic cancer were treated with Gemcitabine and Oxaliplatin combined with oral dosage of Fluorouracil in the Chinese Academy of Medical Sciences Cancer Hospital.Excluded 4 patients,because 2 were not evaluated and 2 patients lost follow-up,27 patients were included in the study.The efficacy and toxicity were evaluated according to the efficacy evaluation criteria(RECIST)1.0 of the solid tumor and the National Cancer Institute toxicity criteria(NCI-CTC)4.0,respectively.Gemcitabine 1000mg/m~2 and Oxaliplatin 85mg/m~2 were administered by intravenous infusion at the first day of chemotherapy;Fluorouracil drugs:Capecitabine 1000mg/m~2 or TS-1 40mg/m~2 were administered twice per day by orally from day 1 to day 8.Every 14 days is a cycle and efficacy was assessed every 4 cycles.Results:Patients who received gemcitabine,oxaliplatin and oral fluorouracil(capecitabine or tiggio),a median progression-free survival(mPFS)and median overall survival(mOS)of 5.91 months(95%CI:2.24?9.59 months)and 11.8 months(95%CI:10.8-12.8 months),respectively;The objective response rate(ORR)was 22.2%;the disease control rate(DCR)was 55.5%,;The grade 3/4 adverse events included neutrophil reduction in 7 cases(25.9%),1 case of diarrhea(3.7%),vomiting in 1 case(3.7%),and 1 case of oral ulcer(3.7%).Conclusion:Gemcitabine and oxaliplatin combined with oral dosage of fluorouracil is effective and well-tolerated in the treatment of locally advanced and metastatic pancreatic cancer.
