Objective: To discuss the relationship between PM2.5 and respiratory system.Through the analysis of the patients of pulmonary disease department outpatient service and pm2.5 meteorological factors,and creating animal models of lung damage by airborne inhalable particles affecting mice and utilize Yupingfeng powder to prevent,prevent and treat as well as treat the animal models.Discuss effects of the damage by PM2.5 and therapy through the analysis of pathology,tunica mucosa trachea and inflammatory factors.Methods:In clinical research,collecting information of PM2.5,PM10 and AQI and basic information of outpatient in respiratory department of Traditional Chinese Medicine of Shan dong Province to observe the correlation between meteorological conditions and outpatient visits at all ages in 2015.In experimental research,divide the mice into five groups as control group,model group,treatment group,prevention + treatment group and prevention group.Infuse smoke produced after burning the coal into the sealed glass box with the inhalation of mice to produce lung injury in mice model.Afterwards,mice were treated with different intervention methods to lavage by Yupingfeng powder.Take the alveolar lavage fluid to detect the number of dust cells.The pathological changes of lung and trachea were observed under light microscope and transmission electron microscope.The expression of CD68 and NF-KB P65 mRNA was detected by PT-PCR.The content of CD4 and CD8 was detected by flow cytometry.The levels of TNF-?,IL-1,IgG,IgE,NF-KB and TRPV1 in serum IgG,IgE and lung tissue homogenate were measured by ELISA.The number of CD68-positive cells was determined by immunohistochemistry.Determination of MDA content and SOD activity in lung homogenate are tested by biochemical method.Results: Clinical studies have shown that meteorological factors are related to the number of patients who have been delayed for 2 weeks with PM2.5 predominantly positive correlated.Experimental results show that(1)dust cells:Simple prevention of intervention did not improve the number of dust cells.But the treatment,treatment + prevention of intervention can reduce the number of dust cells,and the treatment + prevention group is more significant.(2)lung histopathology: After exposure,mice can be seen in the lung tissue filled with inflammatory cell infiltration.In the Yupingfeng powder intervention,we can see in the mouse lung tissue,inflammatory changes are alleviated.And the treatment + prevent group is the best.(3)IgG,IgE in serum and lung tissue: In the treatment + prevention group and prevention group,Yupingfeng powder can significantly reduce the serum IgE content after exposure and can also increase the serum IgG content.(4)Inflammatory cytokines IL-1,TNF-?,NFKB,TRPV1: After exposure,the values were increased.After the intervention of Yupingfeng Powder,the levels of TNF-?,NFKB and TRPV1 were significantly decreased and IL-1 had no obvious change.The effect of improvement in treatment + prevention group was the most obvious.(5)T lymphocytes: CD4,CD8 decreased significantly after exposure.After the intervention of Yupingfeng Powder,in treatment + prevention group and prevention group,the visible value increased.(6)CD68 positive cells: After exposure,the values were increased.After the intervention of Yupingfeng Powder,the values were decreased.The effect of improvement in treatment + prevention group was the most obvious.(7)CD68,NFKB gene expression levels: After exposure,the values were increased.After the intervention of Yupingfeng Powder,the values were decreased.The effect of improvement in treatment + prevention group was the most obvious.(8)MDA?SOD: After exposure,MDA was increased and SOD decreased significantly.After the intervention of Yupingfeng Powder,The effect of improvement in treatment + prevention group was the most obvious.Conclusion: PM2.5 has some damage to lung tissue.In the treatment of traditional Chinese medicine,Yupingfeng powder intervention can play a certain role in improving.And the effect of improvement in treatment + prevention group was the most obvious.Prevention group in front of the canister to the premise,also play a obvious prevention role,after continue to infected index improved significantly in the model group.Thus,Yupingfeng powder can be used to rectify the damage in lung tissue caused by PM2.5,which is meaningful to the development of Chinese medicine.
