Reserch Of Coix Seed And Its Compositions On Tumour Growth Inhibitation

Objective: To investigate the effect of the coix seed and composition on the tumour growth inhibition and the inhibiting effect on A549 and MCF cell in vitro.Methods: Make a preparation of H22 and S180 tumour first and then 70 Kunming mice were randomly assigned to one of seven groups: model group,positive DDP group,and protein of coix seed group,starch of coix seed group,poly saccharide of coix seed group,water decoction group and essential oil of coix seed group.And choose the same number of healthy mice as a normal control group with the amount of administration controlled.The dose of the coix seed is calculated by the equivalent dose coefficient method described in the Chinese Pharmacopoeia.The does is set as twice of the highest dose.The intragastric administration is one time one day and measure the length,radius and weight of the tumour after the operation.After 10 days,the thymus index,spleen index,tumor inhibition rate and tumor volume were calculated from the measured weight of body,tumor,thymus and spleen.Make a preparation of the human lung Aden carcinoma cell A549 and human breast cancer MCF for experiments.The DDP group is set as 8μg/m L,protein of coix seed group is set as4μg/m L,starch of coix seed group is set as 25μg/m L,poly saccharine of coix seed group is set as12.5μg/m L,water decoction group is set as 360μg/m L and essential oil of coix seed group is set as 16μg/m L.By employing different concentrations of coix seed and its composition,the cell proliferation level and the inhibition rate was calculated by MTT assay after 24 and 48 hours.Results:For the H22 tumour,compared with the mode group,the thymus and spleen index significantly decreased in DDP group(P <0.01),the weight of the tumour reduced(P <0.01),the inhibition rate increased(P <0.01),the volume of the tumour decreased(P <0.01),the weight decreased(P <0.01).As for the water decoction and essential oil of coix seed group,the weight of the tumour reduced,the inhibition rate increased and the volume of the tumour decreased.The inhibition effect of the water decoction was worse than essential oil of coix seed group.Compared with the DDP group,the thymus and spleen index and the weight significantly increased(P <0.01)in protein of coix seed group,starch of coix seed group,poly saccharide of coix seed group,water decoction group and essential oil of coix seed group.The effect was oil of coix seed > the water decoction>poly saccharide of coix seed> starch of coix seed > protein of coix seed.For the S180 tumour,compared with the mode group,the thymus and spleen index significantly decreased in DDP group(P <0.01),the weight of the tumour reduced(P <0.01),the inhibition rate increased(P <0.01),the weight decreased(P <0.01).As for the water decoction and essential oil of coix seed group,the weight of the tumour reduced,the inhibition rate increased(P <0.01 or P<0.05)and the volume of the tumour decreased(P<0.01 or P<0.05).The inhibition effect of the water decoction was worse than essential oil of coix seed group.Compared with the DDP group,the thymus and spleen index and the weight significantly increased(P <0.01)in protein of coix seed group,starch of coix seed group,poly saccharide of coix seed group,water decoction group and essential oil of coix seed group.The effect was essential oil of coix seed > poly saccharide of coix seed >the water decoction > starch of coix seed > protein of coix seed.Coix seed and its composition were used to treat A549 cells.Compared with the blank control group,the proliferation of tumour cells was depressed in the oil,DDP group(P<0.01 or P<0.05)after 24 hours.The proliferation of tumour cells was depressed in the water decoction,oil and DDP group(P <0.01)after 48 hours.Coix seed and its composition were used to treat MCF cells.Compared with the blank control group,the proliferation of tumour cells was depressed in the oil,DDP(P <0.01 or P<0.05)after 24 hours.The proliferation of tumour cells was depressed in the poly saccharide and water decoction,oil and DDP group(P <0.01)after 48 hours.Conclusion: The growth of the tumour could be inhibited by the composition of the coix seed,such as the poly saccharide,water decoction,essential oil.The coix seed and its components had a certain protective effect on the immune organs.With the extension of time,coix seed polysaccharide,coix Seed Decoction and essential oil of coix could significantly inhibit the proliferation of A549 and MCF cells.
